摘要 目的 了解人常见冠状病毒NL63、229E、OC43和HKU1核衣壳蛋白(N蛋白)之间的抗原交叉反应特征.方法 对人冠状病毒NL63、229E、OC43和HKU1的N蛋白用大肠杆菌进行表达和纯化,用竞争ELISA方法对已知的人阳性血清进行交叉反应分析.结果 同属内NL663和229E之间及OC43和HKU1 N蛋白之间存在抗原交叉反应.结论 人常见冠状N蛋白抗原交叉反应特征为以后基于人冠状病毒N蛋白的血清学研究和免疫学诊断试剂研发提供了重要的实验依据. Objective To analyze the antigen cross-reactivity among N protein of human coronavir- us NL63, 229E, OCA3 and HKU1. Methods The purified N protein of human eoronaviruses NL63, 229E, OCA3 and HKU1 were expressed in E. coli and purified. The cross-reactivity was assessed using competitive enzyme linked immunosorbent assay (ELISA) with the positive human serum samples against N proteins. Results The antigen cross-reactivity was detected in same coronavirus species between NL63 and 229E, OCA3 and HKU1. Conclusions This findings provide an important basis for human coronavirus seroepdemiological studies and development of immunoassays for coronavirs based on N proteins.
