摘要
目的建立一种新的石墨炉原子吸收法来快速测定全血中的铝。方法向200μl全血样品中加入800μl混合处理液(含40 ml/L硝酸与6 ml/L过氧化氢),静置20 min后,4℃条件下,12000 r/min离心6 min,离心分离脱去全血中的蛋白,以3.0 g/L硝酸镁为化学改进剂,上清液用石墨炉原子吸收光谱法直接测定。结果在铝的最大吸收波长309.3 nm下,血液中铝含量在10μg/L^250μg/L范围内线性良好,相关系数r=0.9998。此方法的最低检出限为3μg/L,加标回收率为92.5%~110.0%,相对标准偏差为(RSD)为1.9%~9.7%。结论方法已成功应用于33例血透病人全血样品中铝的测定,结果令人满意。
Objective To establish a novel method for rapid determination of aluminum in whole blood by graphite furnace atomic absorbance spectrometry( GFAAS). Methods The 200 μl whole blood sample was added with 800 μl mixture of 40 ml/ L nitric acid and 6 ml/ L hydrogen peroxide,and placed for 20 min at 4℃,then centrifugated at the speed of 12000 r/ min for 6 min at 4℃ to remove the protein in whole blood. Then the whole blood aluminum was directly determined in the supernatant by GFAAS with 3. 0 g/ L Mg( NO3)2as matrix modifier. Results At the maximum absorption wavelength of 309. 3 nm,the linear range of aluminum in whole blood was 10 μg/ L - 250 μg/ L( r = 0. 9998),the detection limit was 3 μg/ L,the recovery was 92. 5% - 110. 0%,and the RSD was 1. 9% - 9. 7%. Conclusion The method has been successfully applied to the determination of aluminum in whole blood samples from 33 hemodialysis patients.
