幽门螺杆菌疫苗构建与其免疫产蛋鸡效果评价分析

摘要 目的:构建幽门螺杆菌疫苗,并通过肌肉、皮下多点注射免疫产蛋鸡,观察其产生体液免疫应答水平,评价幽门螺杆菌疫苗对产蛋鸡的免疫功能影响,筛选高效、新型幽门螺杆菌疫苗。方法:幽门螺杆菌国际标准菌株与临床分离的幽门螺杆菌传代扩增,获得混合菌体,分别辅以0.7 mg氢氧化铝佐剂和70μg CpG,或用pcDNA3.1(+)/ureI重组载体转染HeLa细胞等方法制备多种疫苗抗原,通过分析疫苗免疫产蛋鸡产生抗Hp特异卵黄抗体水平,评价疫苗免疫效果。结果:CpG+灭活菌体组与pcDNA3.1(+)/ureI核酸疫苗组免疫产蛋鸡产生抗体的效果好。结论:卵黄中特异性抗体水平受抗原浓度、佐剂、免疫途径、免疫周期等多种因素影响,通过免疫产蛋鸡效果评价,为其高产制备提供实验依据。
