摘要
目的:研究肉苁蓉配伍维生素C对大鼠运动性肾缺血再灌注损伤的保护作用及机制。方法:将6周龄雄性Wistar大鼠96只为对象随机分为6组:对照组(C组,12只)、一般训练组(M组,12只)、过度训练组(OM组,24只)、肉苁蓉+过度训练组(COM组,16只)、维生素C+过度训练组(VCOM组,16只)、肉苁蓉+维生素C+过度训练组(CVCOM组,16只),进行56 d的力竭游泳训练。COM组、CVCOM组采用专业灌胃器灌胃(ig),每天一次,剂量为6.01g·kg-1,ig体积为5 mL·kg-1;VCOM组、CVCOM组采用腹腔注射(ip),剂量为100 mg·kg-1,每天一次,注射体积为0.5 mL,其他各组ig等量生理盐水。末次训练后24 h,进行血清尿素氮、血清肌酐等生化指标检测。结果:1)8周的过度训练导致大鼠运动性肾缺血"再灌注",肾组织超微结构被严重破坏、组织病理学发生明显改变,肾功能受到严重损坏。2)血尿素氮和血清肌酐上升。OM组(P<0.01)、COM组(P<0.01)、VCOM组(P<0.01)、CVCOM组(P<0.05)高于C组;COM组(P<0.05)、VCOM组(P<0.05)、CVCOM组(P<0.01)低于OM组;COM组(P<0.05),CVCOM组(P<0.01)低于VCOM组。3、SOD活性降低。OM组(P<0.01)、COM组(P<0.01)、VCOM组(P<0.01)、CVCOM组(P<0.05)低于C组;COM组(P<0.05)、VCOM组(P<0.05)、CVCOM组(P<0.01)高于OM组;COM组(P<0.05),CVCOM组(P<0.01)高于VCOM组。4、MDA含量升高。OM组(P<0.01)、COM组(P<0.01)、VCOM组(P<0.01)、CVCOM组(P<0.05)高于C组,COM组(P<0.05)、VCOM组(P<0.05)、CVCOM组(P<0.01)低于OM组;COM组(P<0.05),CVCOM组(P<0.01)低于VCOM组。结论:1)过度训练造成了大鼠肾脏缺血再灌注损伤。2)肉苁蓉和维生素C均可以增强SOD活性,清除氧自由基,减轻脂质过氧化作用,对运动性缺血再灌注肾脏具有保护作用。3.肉苁蓉作用优于维生素C,二者配伍使用效果更好。
Objective :To study the Herba Cistanches combined with vitamin C's protective effect and mechanism of movement in rats' renal ischemia - reperfusion injury. Methods : Randomly divide 96 7 - week - old male Wistar rats into 6 groups : control group ( group C, 12), general training group (group M, 12), overtraining group (group OM, 24), cistanche + overtraining group (group COM, 16 ) , vitamin C + overtraining group (group VCOM, 16 ), and cistanche + vitamin C + overtraining group ( group CVCOM, 16 ) , take a 56 - day of exhaustive swimming training. The COM group, CVCOM group with professional gavage for gastric lavage (ig) , once a day, the dose of 6.01 g·kg^-1Ig volume 5mL. kg^-1 ; group VCOM, group CVCOM ip take intraperitoneal injection, 100 mg·kg^-1 once a day, the injection volume is 0. 5 mL;other groups ig normal saline. Detect biochemical parameters of serum urea nitrogen and serum creatinine 24 hours later after the last training. Results : 1 ) In the 8 week of overtraining, rats resulted in movement of "renal ischemia reperfusion", the ultrastructure of kidney was severely damaged, the histopathological changes obviously, renal function was seriously damaged. 2)Serum creatinine and blood urea nitrogen rise. OM group (P〈0.01), COM group (P〈0.01), VCOM group (P〈0.01), CVCOM group (P〈0.05) higher than in group C ; COM group ( P 〈 0. 05 ), VCOM group ( P 〈 0. 05 ), CVCOM group ( P 〈 0.01 ) lower than the OM group ; COM group(P 〈 0.05 ), CVCOM groups(P 〈 0.01 ) lower than the VCOM group. 3) SOD activity decreased. OM group (P 〈 0. 01 ), COM group (P 〈 0.01 ), of VCOM group (P 〈 0.01 ), CVCOM group (P 〈 0. 05 ) lower than in group C ; M group ( P 〈 0. 05 ), COM group(P 〈0.05), VCOM group(P 〈0.05), CVCOM group(P 〈0.01 ) higher than the OM group; COM group(P 〈0. 05), CVCOM group(P 〈0. 01 ) higher than the VCOM group. 4)The content of MDA increased, OM group ( P 〈 0.01 ), COM group ( P 〈 0. 01 ), VCOM group ( P 〈 0. 01 ), CVCOM group(P 〈0.05) higher than in group C;COM group(P 〈0.05), VCOM group (P 〈 0. 05 ), CVCOM group ( P 〈 0. 01 ) lower than the OM group ; COM group ( P 〈 0.05 ), CVCOM groups ( P 〈 0.01 ) lower than the VCOM group. Conclusion : 1 ) Overtraining can induce the renal ischemia -reperfusion in rats. 2)Cistanche deserticola Y. C. Ma and vitamin C can enhance the activity of SOD, scavenging oxygen free radicals, reduce lipid peroxidation, they have a protective effect on exercise - induced ischemia - reperfusion kidney. 3 ) Cistanche is better than vitamin C, but they worked even better when taken both.
