摘要
本文对1株从土壤中新发现的链霉菌S015中的吩嗪类活性产物进行了结构鉴定及发酵优化。首先,通过16SrDNA鉴定S015为洛蒙德链霉菌。其次,通过2-丁酮萃取及制备型HPLC,在S015的发酵液中纯化得到1个活性产物,并由LC-MS和H-NMR,鉴定该产物为6-醛基-4,7,9-三羟基-吩嗪-1-甲酸酯,即洛蒙真菌素。最后,通过发酵优化,在YE培养基中添加0.1mmol/L的FeCl3,使洛蒙真菌素的产量由(20.6±4.6)mg/L提高到(136.2±18.1)mg/L,为其工业化应用奠定了基础。
An antibiotic phenazine derivative was studied in a new Streptomyces strain S015 isolated from the soil around Shanghai.Strain S015 was confirmed as Streptomyces lomondensis by 16S rDNA nucleotide analysis.One bioactive compound was isolated and purified from the culture broth of S.lomondensis S015 through 2-butanone extraction and preparative HPLC.The compound was identified as a modified phenazine,lomofungin,using LC-MS and H-NMR analysis.Through medium optimization,lomofungin production in S.lomondensis S015 increased from (20.6 ± 4.6) mg/L to (136.2± 18.1) mg/L with the addition of 0.1 mmol/L FeCl3.The information obtained in this work is considered useful for large-scale production of phenazine products.
