清胰化积方在晚期胰腺癌治疗中可抑制MMP-2表达 被引量:3

摘要 目的:研究清胰化积方干预后MMPs的表达变化对胰腺癌肝转移的影响。方法:通过脾脏注射法建立裸鼠人胰腺癌SW1990HM肝转移模型,观察清胰化积方干预后对胰腺癌肝转移灶增殖的影响,通过检测MMPs mRNA表达的变化观察其与肝转移灶之间的关系,并通过免疫组化和western blot进一步验证。结果:①与NS组相比,QYHJ各组均可抑制肝转移灶的增殖生长(P<0.05);②与NS组比较,QYHJ干预后,大、中、小剂量组的肝转移灶肿瘤组织中MMP-1、MMP-7、MMP-9的mRNA表达无明显差异(VS NS,P>0.05);MMP-2的mRNA表达下调(VS NS,P<0.05)。结论:清胰化积方可能通过诱导MMP-2的表达下调抑制胰腺癌肝转移的发展。 Objective:To study the effects of Qingyi on expression of MMPs in pancreatic cancer for liver metastasis. Methods:To building human pancreatic cancer SW1990HM liver metastasis model in nude mice by injection of spleen, observing effects of the Qinyi Huaji recipe in liver metastases proliferation from pancreatic cancer, by detecting changes in the expression of MMPs mRNA and verified by immunohistochemistry and western blot. Results:I. Compared with NS group, QYHJ group can inhibit the proliferation of the growth of liver metastases (P〈0.05); II. Compared with NS, with QYHJ intervention, in large liver metastases tumor tissue, small-dose group, MMP-1, MMP-7, mRNA MMP-9 expression were no significant difference (VS NS, P〉0.05);mRNA MMP-2 is downregulated (VS NS, P 〈0.05). Conclusion:QYHJ party can induce the expression of MMP-2 downregulation to inhibit the development of pancreatic cancer liver metastasis.
