Association of interleukin 10 gene promoter polymorphisms and obsessivecompulsive disorder

摘要 目的探讨白细胞介素-10(IL-10)基因启动子多态性与强迫症(OCD)的关系。方法381例OCD患者和485名健康对照为研究对象,通过聚合酶链式反应与限制性片段长度多态性(PCR—RFLP)基因分型技术对IL-10基因标签单核酸多态性(SYP)位点592A/C(rs1800872)、1082A/G(rs1800896)、819T/C(rs1800871)进行基因分型;以耶鲁一布朗强迫量表(Y-BOCS)评定OCD患者的病情。结果OCD组和对照组IL-10基因rs1800872、rs1800896、rs1800871位点在基因型频率分布与等位基因频率分布差异均无统计学意义(P〉0.05);不同发病年龄女性3个SNPsOCD组与对照组人群基因型与等位基因频率分布无明显差异(P〉0.05);男性rs1800896基因型(X2=6.51,P=0.039)和等位基因(X2=6.59,P=0.01,OR=1.84,95%CI=1.15—2.95)频率分布在OCD组与对照组之间存在差异;OCD组男女性别之间rs1800896基因型(矿=8.08,P=0.02)和等位基因(X2==6.97,P=0.008,OR:1.95,95%CI:1.18—3.23)频率分布也存在差异。结论IL-10基因(rs1800896)可能与男性OCD易感性有关。 Objective To investigate the association between interleukin 10 (IL-10)gene promoter polymorphism and obessive-compulsive disorder(OCD). Methods Genotyping for IL-10 gene SNP592A/C ( rs1800872), 1082 A/G (rs1800896) ,and 819T/C( rs1800871 )were performed for 381 OCD patients and 485 health controls with polymerase chain reaction and restriction fragment length polymorphism techniques. The OCD patients were assessed with Yale- Brown Obsessive-Compulsive Scale (Y-BOCS). Results There were no significant differences in the frequency distribu- tion of genotype and allele of IL-10 gene( rs1800872 ,rs1800896 ,and rs1800871 )between OCD patients and health con- trols( P 〉 0. 05 ). There were no significant differences in the freqency distribution of genotype and allele of 3 single nu- cleotide polymorphisms (SNPs) for the females of different onset age between OCD group and control group( P 〉 0. 05 ). There were significant differences in the freqency distribution of genotype (X2 = 6. 51, P = 0. 039 ) and allele (X2 = 6. 59, P = 0.01 ; odds ratio [ OR ] = 1.84,95 % confidence interval [ 95 % CI ] = 1.15 - 2.95 ) of IL- 10 gene ( rs 1800896 ) for the males between OCD patients and health controls ,and there were significant differences in the freqency distribution of geno type(x2 =8.08,P =0.02)and allele(x2 =6.97,P = 0.008;OR = 1.95,95% CI= 1.18 -3.23)of IL-10 gene (rs1800896)between female OCD group and male OCD group. Conclusion The results support a role for IL-10 gene (rs1800896) in conferring susceptibility to OCD in male.
