摘要
目的探讨黄芪甲苷对卡介苗(BCG)和脂多糖(LPS)诱导肝损伤的保护作用。方法 60只SD大鼠随机分为对照组,模型组(BCG+LPS),黄芪甲苷(20、40、80 mg/kg)组和联苯双酯(150 mg/kg)组,采用BCG2.5 mg/只静脉注射致敏大鼠,第10 d给予LPS 75μg/只静脉攻击,复制大鼠免疫性肝损伤模型,黄芪甲苷或联苯双酯组分别同时灌胃给予黄芪甲苷或联苯双酯,连续10 d,分光光度法测定大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST),超氧化物歧化酶(SOD),丙二醛(MDA)含量;观察肝组织形态学变化;称重法测定肝脏、脾脏指数;酶联免疫吸附法测定血清中TNF-α,IL-6水平。结果与模型组比较,40、80 mg/kg黄芪甲苷组大鼠血清AST、ALT活性降低、肝脏、脾脏指数降低;与对照组比较,模型组大鼠血清中TNF-α[(60±4.2)pg/mL]、IL-6[(289±12.8)pg/mL]水平、MDA含量[(25.3±2.8)nmol/mg prot]升高,SOD活性[(72.2±9.8)U/mg prot]下降;与模型组比较,80 mg/kg黄芪甲苷组大鼠血清中TNF-α[(26±1.5)pg/mL]、IL-6[(139±13.8)pg/mL]水平、MDA含量[(14.2±2.9)nmol/mg prot]明显下降,SOD活性[(108.1±8.3)U/mg prot]升高。结论黄芪甲苷对大鼠免疫性肝损伤具有一定保护作用。
Objective To study protective effect of astragaloside IV on immunological liver injury induced by bacillus Calmette-Guerin(BCG) and lipopolysaccharide ( LPS ). Methods Sixty Sprague-Dawley (SD) rats were randomly divided into 6 groups: normal group, model group, different dose of astragaloside IV groups ( 20,40,80 mg/kg), and bi- phenyl positive control group (150 mg/kg). The model of rat immunological liver injury was induced by injecting 2. 5 mg/rat BCG and 75 μg/mt LPS at the tenth day. The content of alanine transaminase ( ALT), aspirate aminotrans- ferase ( AST ), superoxide dismutase ( SOD ), and malondialdehyde ( MDA ) were measured by spectrophotometric method. Liver pathological changes were observed with hematoxylin-eosin (HE) staining. The liver index and spleen in- dex were determined by weighting method. The content of tumor necrosis factor ct ( TNF-ct ) and interleukin-6 ( IL-6 ) were determined with enzyme-linked immunosorbent assay(ELISA). Results Compared with the model group, the ac- tivity of ALT,AST and the liver and spleen index were decreased in astragaloside IV(40,80 mg/kg) groups. Compared with the control group,the content of TNF-a(60 ±4. 2 pg/mL) ,IL-6(289 ± 12. 8 pg/mL) and MDA(25.3 ±2.8 nmol/ mg prot)increased and the activity of SOD (72. 2 ± 9.8 U/mg prot)decreased in model group. Compared with the model group,the content of TNF-ct (26 ± 1.5 pg/mL) ,1L-6 ( 139 ± 13.8 pg/mL) and MDA( 14. 2 ± 2. 9 nmol/mg prot) de- creased significantly,and the SOD activity( 108.1± 8.3 U/mg prot)increased significantly in astragaloside IV( 80 mg/ kg) group. Conclusion Astragaloside IV has a protective effect on immunological liver injury.
