莪术散对子宫内膜异位症模型大鼠异位内膜组织Cyclin A及P21表达的影响 被引量:3

摘要 目的:探讨子宫内膜异位症(内异症)大鼠在位和异位组织中细胞周期蛋白A(Cyclin A)及P21表达的差异,从细胞周期调控的角度探讨莪术散治疗子宫内膜异位症的相关机制,以及对其调节作用。方法:用自体移植法建立内异症大鼠模型,将造模成功的大鼠随机分成复方莪术散高、低剂量组,孕三烯酮对照组,模型对照组和空白对照组,共5组,以1 mL/100 g灌胃给药。采用免疫组化法测定给药后大鼠在位以及异位内膜组织中Cyclin A及P21的表达。结果:Cyclin A在子宫内膜异位症组中其阳性表达率较其在正常子宫内膜组织中增高,P21蛋白在子宫内膜异位症组中其阳性表达率较其在正常子宫内膜组织中明显降低,Cyclin A与P21蛋白在子宫内膜异位症组中的表达呈负相关;复方莪术散在一定剂量关系上能显著抑制异位组织的生长,缩小异位病灶体积,抑制Cyclin A的表达,提高P21蛋白的表达,与模型对照组比较有统计学差异(P<0.05)。结论:莪术散可抑制Cyclin A的表达,提高P21蛋白的表达,从细胞周期调控的角度证实复方莪术散对子宫内膜异位症具有调节作用,这可能是其治疗子宫内膜异位症的机制之一。 Objective:To observe the differences of Cyclin A and P21 expressions in eutopic and ectopic tissues of rats with endometriosis and explore the mechanism of compound Ezhtt powder to treat endometrio- sis from the aspect of the cell cycle regulation. Methods :The rat model with endometriosis was established with self-endometrium transplantation. The established rat models were divided into five groups:compound Ezhu power high dosage group,compound Ezhu low dosage group,gestrinone control group,model control group,and the normal control group, given the garage of 1 mL/100 g. After the treatment,the expressions of Cyclin A and P21 of in eutopic and ectopic tissues were tested with immuno-histochemistry method. Re- suits:The expression of Cyclin A in endometriotic tissues was significantly higher than that in normal en- dometrial tissues,whereas the expression of P21 in endometriotic tissues was significantly lower than that in normal endometrial tissues. There was negative correlation between expression of P21 and Cyclin A in en- dometriosis. Compound Ezhu power inhibited the growth of endometriotic tissue in rat model,decreased the volume of ectopic focus,suppressed expression of Cyclin A,and enhanced the expression of P21,which obvi- ous statistical difference compared with the model group(P〈0.05). Conclusion:Compound Ezhu power could inhibit the expression of Cyclin A,and increase the expression of P21. From the aspect of cell cycle regula- tion,it is confirmed that the compound Ezhu power has a regulatory effect on endometriosis,and such regulatory effectmay be one of the mechanism to treat endometriosis.
