不同剂量X线照射EC9706细胞对NADH脱氢酶活性和6Gy照射对mtDNA复合物Ⅰ基因序列的影响 被引量:1

摘要 目的探讨不同剂量X线照射食管鳞癌Ec9706细胞对NADH脱氢酶活性的影响以及6GyX线照射对线粒体DNA(mitochondrial DNA,mtDNA)复合物Ⅰ基因序列的影响。方法①通过2、4、6、8Gy剂量X线照射食管鳞癌Ec9706细胞,48 h后收集被照射细胞,提取线粒体,测试酶活性。②6Gy剂量X线照射Ec9706细胞后,提取mtDNA,经PCR扩增后,纯化PCR产物,测Ec9706细胞mtDNA复合物Ⅰ基因序列,并进行对比。结果①据统计结果分析:2、4、6、8Gy剂量照射食管鳞癌Ec9706细胞后,各组之间酶活性有显著性差异(P<0.05),6Gy组酶活性显著低于其它四组(P<0.01)。②6GyX线照射前后Ec9706细胞mtDNA复合物Ⅰ基因序列没有变化。结论①经2Gy、4Gy、6Gy、8Gy X线照射后,Ec9706细胞NADH脱氢酶活性都低于未照射组,从2Gy到6Gy,随着剂量增加酶活性逐渐降低,在6Gy剂量时降到最低;当剂量逐渐增大到8Gy,酶活性虽然低于对照组,但不再继续降低。②6GyX线照射对Ec9706细胞mtDNA复合物Ⅰ基因序列没有造成影响。 Objective To explorethe effect of different doses of X - ray radiation in esophageal squamous cell Ec9706 NADH dehydrogenase activity and 6Gy - X - ray radiation on mitochondrial DNA(mtDNA) I gene sequences comple- xes. Methods (1) By 2,4,6,8 Gy dose of X - ray radiation in esophageal squamous cell Ec9706 cells,48 h after irradi- ation cells were collected, extracted mitochondrial testing activity. (2) After the X - ray radiation dose 6Gy Ec9706 ceils, extracted mtDNA,after PCR amplification, PCR products were purified, mtDNA I gene sequences composite measure Ec9706 cells and compared. Results (1) According to statistics analysis : Ec9706 esophageal squamous cell carcinoma ceils after irradiation with 2,4,6,8 Gy dose of activity between the groups was significant difference (P 〈 0. 05 ), 6Gy group activity was significantly lower than the other four groups( P 〈 0. 01 ). (2) MtDNA I gene sequences Ec9706 cell complexes line did not change before and after radiation 6GyX. Conclusion (1) After 2Gy,4Gy,6Gy,8Gy X - ray radi- ation after, Ec9706 cell NADH dehydrogenase activity was lower than the non - irradiated group, from 2Gy to 6Gy, activi- ty as the dose increased gradually reduced to a minimum when 6Gy dose;When the dose was gradually increased to 8Gy, although activity than the control group, but no longer continue to reduce. (2) 6GyX irradiation on mtDNA I gene se- quences complexes Ec9706 cells are not affect
