BM-cyclin1上调转录因子Sp1促进TopoⅡα表达逆转肿瘤多药耐药的研究

摘要 目的:探讨BM-cyclin 1逆转人口腔上皮癌C-A120细胞多药耐药的机制。方法:MTT法检测细胞增殖;RT-PCR及Western blot检测BM-cyclin 1对TopoⅡαmRNA、蛋白表达的影响,Western blot检测BMcyclin 1对转录因子Sp1、Sp3表达的影响;报告基因法检测BM-cyclin 1对Sp1、Sp3转录调控的影响。结果:(1)BM-cyclin 1可以明显逆转C-A120细胞多药耐药:C-A120细胞对ADM、VP-16、DDP敏感性分别提高了6.0、8.2及1.7倍。(2)BM-cyclin 1明显上调TopoⅡαmRNA及蛋白表达上调。(3)BM-cyclin 1可明显诱导转录因子SP1表达并上调其转录活性。结论:BM-cyclin 1能够明显逆转C-A120细胞多药耐药性;并且这种逆转作用可能与BM-cyclin 1上调转录因子Sp1活性,增加TopoⅡα表达相关,为BM-cyclin 1进一步开发应用提供理论依据。
