硫化氢对大鼠肝硬化及门静脉高压的保护作用 被引量:1

摘要 目的观察硫化氢在减轻CCl4造成的肝硬化和门静脉高压中的作用。方法雄性成年大鼠42只随机分为实验组36只,对照组6只。其中,实验组随机分为盐水组、硫氢化钠组和DL-炔丙基甘氨酸(PAG)组,每组各12只,分别腹腔注射1ml生理盐水、硫氢化钠液体(10μmol/kg)和PAG液体(30mg/kg),每2d1次,且实验组大鼠都给予腹腔注射50%CCl4(1ml/kg,2次/周),对照组给予等量生理盐水腹腔注射,12周后检测肝功、肝纤维化指标、肝组织病理等。结果硫氢化钠组丙氨酸转氨酶(AIJT)、天冬氨酸转氨酶(AST)、透明质酸(HA)、肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、向细胞介素(IL)-6、细胞间黏附分子(ICAM)-1、肝脏羟脯氨酸、α-平滑肌肌动蛋白(d—SMA)表达量分别为(137.6±20.3)U/L、(317.5±78.2)U/L、(296.3±32.1)μg/L、(109.3±32.1)ng/L、(50.6±9.8)ng/L、(216.4±42.1)ng/L、(153.6±22.8)ng/L、(616.8±138.5)μg/g、0.53±0.14,均低于盐水组(P〈0.05);硫氢化钠组血清白蛋白为(4.4±0.8)g/dl,高于盐水组(P〈0.05);硫氢化钠组门静脉的压力为(13.78±3.84)cmH2O(1cmH2O=0.098kPa),低于盐水组(P〈0.05);但两组肝脏表达胱硫醚-1-裂解酶(CSE)和脾脏重量差异无统计学意义P〉0.05。结论外源性硫化氢可减轻CCl4造成的肝硬化和门静脉高压。 Objective To investigate the role of H2 S in carbon tetrachloride ( CCl4 ) -induced cirrhosis and portal hypertension. Methods 42 rats were divied into the experimental group (n =36) and control group (n = 6). The experimental group consisted of 3 subgroups (each had 12 rats) : saline, NariS and DLpropargylglycine (PAG), receiving intraperitoneal injections of 1 ml of saline, NariS solution ( 10 mmoL/kg) and PAG solution (30 mg/kg) respeelively. All of the 3 subgroups were given injections of 50% CC14 ( 1 ml/kg, twice a week). The control group were given intraperitoneal injections of equal quantity saline. After 12 weeks, the indices for liver fibrosis and liver pathology were assessed. Results The contents of alanine aminotransferase (ALT), aspartate aminotransferase (AST), hyaluronie acid (HA), proinflammatory cytokines including tumor necrosis factor (TNF)-α, interleukin (IL)-113,IL-6, intercellular adhesionmol- ecule (ICAM)-I, hydroxyproline,α-smooth muscle actin (ct-SMA) in Naris group were (137.6 ± 20. 3) U/L, (317.5 ±78.2) U/L, (296. 3 ±32. 1) μg/L, (109. 3 ±32. 1) ng/L, (50. 6 ±9. 8) ng/L, ( 216. 4 ± 42. 1 ) ng/L, ( 153.6 ± 22. 8 ) ng/L, (616. 8 ± 138.5 ) μg/g, 0. 53 e 0. 14 respectively, significantly lower than in saline group ( P 〈 0. 05 ) ; the albumin of NariS group which was ( 4. 4 ± 0. 8 ) g/dl was higher than saline group ( P 〈 0. 05 ) ; the portal pressure of NariS group which was ( 13.78 ± 3.84 ) cmH20 ( 1 emil20 = 0. 098 kPa) was lower than saline group ( P 〈 0. 05 ) ; There were' nt significant difference between the two groups of the expression of eystathionine -lyase ( CSE ) and the spleen weight ( P 〉 0. 05 ). Conclusion Exogenous H2 S attenuates CCl4-induced liver cirrhosis and portal hypertension.
