Effect of omega-3 polyunsaturated fatty acids on the growth of prostate cancer cells

摘要 目的探讨Omega-3多不饱和脂肪酸对前列腺癌细胞生长的作用和机制。方法通过噻唑蓝(MTT)比色法观察浓度分别为20、40、60、80μmol/L的二十二碳六烯酸对前列腺癌LNCap细胞活力的影响;使用40μmol/L二:十二碳六烯酸处理前列腺癌LNCap细胞0、12、24、36h,通过实时荧光定量聚合酶链反应(FQ.PCR)和Western blot法检测二十二碳六烯酸对LNCap细胞雄激素受体(AR)的影响。结果MTT实验结果显示20、40、60、80μmol/L的二十二碳六烯酸对LNCap细胞增殖的抑制率分别为13.3%、46.7%、53.3%、63.3%,Western blot结果显示40μmol/L的二十二碳六烯酸能够使AR的蛋白表达量下降60%~80%。结论Omega-3多不饱和脂肪酸抑制前列腺癌细胞的生长与其下调AR的表达有关。 Objective To discuss the effect of omega-3 polyunsaturated fatty acids on the growth of prostate cancer cells and the possible action mechanism. Methods The cytotoxic effect of different concentrations of docosahexenoic acid (20, 40, 60 and 80 μmol/L) on prostate cancer cell line LNCap was examined by methyl thiazol tetrazolium (MTT) assay. The mRNA and protein expression level of androgen receptor was detected by real-time fluorescence quantitative polymerase chain reaction (FQ-PCR) and Western Blotting in order to demonstrate the effect of 40 μmol/L docosahexenoic acid on the growth of LNCap cells for 0, 12, 24 and 36 h. Results MTF assay showed that the inhibition ratio of 20, 40, 60 and 80μmol/L docosahexenoic acid on the growth of LNCap cells was 13.3%, 46. 7% , 53.3% and 63.3% respectively. Western blotting revealed that the androgen receptor protein quantity was obviously downregulated with docosahexenoic acid by 60%-80%. Conclusion Omega-3 Polyunsaturated fatty acids inhibit the growth of prostate cancer cells, which may be associated with down-regulation of androgen receptor protein level.
