摘要
目的观察Dicer基因在甲状腺乳头状癌(PTC)中的表达,探讨Dicer基因表达与PTC的发生发展的关系。方法采用免疫组织化学链菌素抗生物素蛋白一过氧化物酶(SP)法和荧光定量逆转录聚合酶链反应技术检测Dicer在50例PTC及其配对的癌旁组织(NCE)的蛋白及mRNA的表达。结果DicermRNA在PTC和NCE中的阳性表达率分别为80%(40/50),10%(550),其在PTC中的表达显著高于NCE(P〈0.01)。Dicer蛋白在PTC和NCE中的阳性表达率分别为76%(38/50),8%(4/50),其在PTC中的表达明显高于NCE(P〈0.01)。Dicer蛋白及其mRNA在PTC中的表达与患者的TNM分期及淋巴转移有关(P〈0.05)。Dicer蛋白和DicermRNA在PTC中的表达有明显的相关性(P〈0.01)。结论Dicer在PTC发生发展中起到重要作用。
Objective To investigate the Dicer gene expression and its clinicopathological signifi- cance in papillary thyroid carcinoma, and the relationship between the Dicer gene expression and tumorogenesis of papillary thyroid carcinoma. Methods The immunohistochemical streptavidin avidin-peroxidase (SP) technique and real-time quantitative polymerase chain reaction ( Real-time PCR) were used to detect the expression of Dicer protein and mRNA in 50 cases of PTC and their matched adjacent non-cancerous epithelia (NCE). Results The positive expression rate of Dicer mRNA in PTC and NCE wase 80% (40/50) and 10% (5/50) respectively (P 〈0. 01 ). The positive expression rate of Dicer protein in PTC and NCE was 76% (38/50) and 8% (4/50) respectively (P 〈 0. 01 ). The expression of Dicer protein and mRNA was associated with the invasion and metastasis of PTC ( P 〈 0. 05 ). The expression of Dicer protein and mRNA showed a distinct correlation ( P 〈 0. 01 ). Conclusion Dicer plays a pivotal role in the carcinogenesis and progression of PTC. Dicer could be a new diagnostic/therapeutic target in PTC.
