姜黄素对视网膜缺血再灌注损伤Bcl-2,Bax表达的影响 被引量:1

摘要 目的研究姜黄素(Cur)预处理对视网膜缺血再灌注损伤(RIRI)Bcl-2和Bax表达的影响,为姜黄素治疗视网膜缺血再灌注损伤(RIRI)提供实验依据。方法将33只健康成熟无眼疾的SD大鼠随机分为正常组(3组)、模型组(15只)、Cur组(15只),缺血前1h,正常组不进行任何处理,模型组腹腔注射生理盐水,Cur组腹腔注射姜黄素,前房加压法建立视网膜缺血再灌注损伤(RIRI)模型,模型组和Cur组分别在2h,6h,12h,24h,48h 5个时间段,通过腹腔注射10g/L戊巴比妥钠处死大鼠,立即摘取实验眼球,通过免疫组化检测Bcl-2和Bax阳性表达。结果正常组未见Bal-2阳性表达,模型组和Cur组在缺血再灌注后2h可见Bal-2阳性表达,6h,12h Bal-2阳性表达逐渐增加,24h阳性表达达高峰,48h阳性表达逐渐减少;Bax在正常组未见阳性表达,模型组和Cur组在缺血再灌注后2h可见Bax阳性表达,6h Bax阳性表达逐渐增加,12h阳性表达达高峰,24h阳性表达逐渐减少,48h表达明显下降;在各时间段Cur组Bcl-2阳性表达数均高于模型组;Bax阳性表达数均低于模型组,各时间段差异具有统计学意义(P<0.01)。结论姜黄素预处理大鼠视网膜缺血再灌注损伤,可增加神经节细胞Bcl-2表达,减少Bax阳性表达,减少神经节细胞损伤,对视网膜缺血再灌注损伤具有保护作用。 Objective To explore the efect of Curcumin on the expressions of Bcl-2 and Bax after retinal ischemia reperfusion in-jury,to provide experimental basis for the treatment of RIRI .Methods A total of 33 healthy mature without eye disease of SD rats were ran-domly divided into three groups:normal group ,Model group ,Curcumin group .One hour before ischemia ,normal group without any treatment , the model group were injected with normal saline solution in the abdominal cavity and the Curcumin group were injected with Curcumin ,Reti-nal ischemia reperfusion injury ( RIRI) model was established with anterior chamber pressure .Model group and Curcumin group rats were kill-ed by intraperitoneal injection of 10g/L sodium in 2h,6h,12h,24h,48h five period,respectively,and immediate removal of experimental eyes.The positive expression of Bcl-2 and Bax were detected by immunohistochemistry .Results There were no positive Bcl-2 expression in normal group .There was positive Bcl-2 expression in model group and Curcumin group 2 hour after ischemia reperfusion ,expression gradually increasing at the 6hour and 12hour and reaching peak at the 24 hour,and expression gradually declining at the 48 hour.There was no positive Bax expression in normal group ,and there was positive Bcl-2 expression in model group and Curcumin group 2 hour after ischemia reperfu-sion,expression gradually increasing at the 6hour,and reaching peak at the 12hour,and expression gradually declining at the 24hour and 48hour.Compared with the model group ,the number of Bcl-2 positive expression at each time point in Curcumin group increased significantly , and the number of Bax positive expression at each time point in Curcumin group declined significantly .There was statistical significance in the expression of Bcl-2 and Bax between the model group and Curcumin group in 2~48 hours after reperfusion .Conclusion Curcumin pretreat-ment in the rat retina Ischemia reperfusion injury ,can increase the ganglion cells the Bcl-2 expression ,reduce Bax positive expression and re-duce ganglion cell damage .The retina ischemia-reperfusion injury has a protective effect and has protective effect on RIRI .
