哮喘小鼠模型肺组织、肺泡灌洗液中TNF-α及IL-12水平比较 被引量:3

Compare between Lung Tissue,TNF-α and IL-12 Levels in Bronchoalveolar Lavage Fluid of Asthmatic Mice

摘要 目的观察支气管哮喘小鼠模型肺泡灌洗液(BALF)中TNF-α,IL-12水平,了解两者的水平及诊断价值。方法 40只雌性Balb/C小鼠,随机分成2组:A正常对照组、B支气管哮喘模型组,每组各20只。采用卵蛋白(OVA)等致敏支气管哮喘模型组;第21天后,哮喘模型组1%卵蛋白溶液雾化吸入,雾化时间共5周,对照组用生理盐水雾化吸入。分别观察两组哮喘发作症状、ELISA检测肺泡灌洗液中TNF-α,IL-12的水平。结果支气管哮喘模型组肺泡灌洗液中TNF-α显著升高,IL-12降低,且IL-12与TNF-α呈密切负相关(r=-0.964,P<0.01)。结论哮喘的发生发展不仅与炎症介质TNF-α的异常释放增多有关,还与内源性的抗炎介质IL-12的释放不足有关,而且IL-12能够抑制TNF-α的合成。 Objective To observe the bronchial asthma model in mice ,understand the level of tumor necro-sis factor-α(TNF-α) and Interleukin-8(IL-8) in bronchoalveolar lavage fluid(BALF),to assess their diagnostic value. Methods Forty female Balb/C mice were randomly divide into normal control group A ( n =20 ) and asthma model group B(n=20) established by ovalbumin(OVA).After the 21th days,challenged with repeated exposure to aerosolized OVA for five weeks in model groups ,and control groups with saline inhalation .Respectively to observe the two groups of asthma symptoms ,BALF were tested in ELISA .Results The level of TNF-αin BALF of asthma model groups was signif-icantly increased,IL-12 was reduced,and IL-12 was closely negative correlation with TNF-α(r=-0.964,P〈0.01). Conclusion The occurrence and development of asthma are not only with the increase in the number of abnormal re -lease of TNF-αinflammatory mediators,also with the release of endogenous anti-inflammatory mediators of IL -12 defi-ciency,and IL-12 can inhibit the synthesis of TNF-α.
