人脐动脉的抗压力性及其用于兔血管移植的研究

摘要 目的探讨人脐动脉的耐压性能,观察辐照氟银脐动脉移植至中国白兔颈动脉后的内膜增生及ki-67的表达。方法液压扩张法检测脐动脉的耐压性能。30只中国白兔的左、右侧颈动脉同时分别植入辐照氟银脐动脉(实验组)和自体颈动脉(对照组),术后2、6周观察血管通畅情况,分别取材作组织形态学观察和免疫组织化学检测,观察血管内膜厚度及血管平滑肌细胞中ki-67的阳性表达。结果人脐动脉具有良好的耐压性能。移植术后2、6周2组兔移植血管内膜厚度比较差异无统计学意义(P>0.05)。实验组兔血管平滑肌细胞中ki-67的表达较对照组高,但2组术后2、6周比较,差异无统计学意义(P>0.05)。结论人脐动脉从耐压性能上具备作为冠状动脉搭桥血管材料的基本条件。辐照氟银脐动脉的内膜增生与自体动脉无明显差异。 Objective To explore the withstand voltage characteristic of human umbilical artery and to investigate the expression of ki-67 and the intimal hyperplasia(IH) of the irradiated human umbilical artery incorporated with norfloxacin and silver(IHUAINS) grafts implanted into the carotid arteries of the rabbit. Methods Hydraulic expansion test was performed on neonatal umbilical arteries. Thirty rabbits were performed bilateral carotid bypass grafting. The IHUAINS( experimental group) and the left carotid arteries( control group)were implanted in the left and right carotid arteries respectively. Graft patency was checked at two weeks and six weeks after implantation, and the grafts were observed by using standard histological techniques and immunohistochemieal method for measurement of intimal thickness and the expression of ki-67. Results Neonatal umbili- cal cord had good withstand voltage characteristic. The difference of the intimal thickness between the two groups at two and six weeks after grafting had no statistical significance (P 〉 0.05 ). At the same time, immunoeytoehemical analysis detected that ex- pression of ki-67 in the experimental group was stronger than that in the control group,there was no significant difference be- tween the two groups at two weeks and six weeks after implantation ( P 〉 0.05 ). Conclusions Umbilical artery may be an ide- al graft because of good withstand voltage characteristic in the field of coronary surgery. Animal experiments showed that IH be- tween IHUAINS and autologous artery has no obvious difference.
