摘要
目的:用脑内注射胶原酶方法建立小鼠脑出血模型。方法:在动物脑立体定位仪下将不同剂量Ⅳ型胶原酶0.5μl注射到昆明小鼠左侧纹状体处,分别在术后24h对小鼠运动功能进行检测,并对出血部位脑组织进行尼氏染色,光镜观察染色结果来验证模型制作是否成功。结果:术后小鼠即表现右侧肢体运动功能障碍。形态学观察证实左侧纹状体内血肿形成,且在一定范围内,出血灶大小与肢体偏瘫症状发展过程基本一致。不同个体间相同单位的胶原酶出现的血肿体积和部位基本一致。结论:应用Ⅳ型胶原酶脑内注射方法建立的小鼠脑出血模型,是一种方法简单,效果可靠,易于大批量制作的动物模型。脑内注射0.02U的IV型胶原酶制备的脑出血模型能较好满足实验要求。
Objective:To establish a typical mouse model of intracerebral hemorrhage (ICH) .Methods :Intracerebral hemorrhage in mice were generated by stereotaxically injecting different dose of collagenase type Ⅳ into the left corpus striatum .Every mouse received a volume of 0 .05μl collagenase type Ⅳ .Neurological scores were as-sessed and recorded 24 hours after injection .After behavioral testing the brains were processed for routine histologi-cal examination .Light microscope observation to determine if the animal model was successful .Results:Mice showed the motor dysfunction after operation .Morphology confirmed that the hematoma formed at the left corpus striatum ,and within a certain range ,the size of hemorrhage and the development of symptoms of hemiplegia was re-lated .Different individuals of the same unit of collagenase type Ⅳmaking the volume and location of hematoma was same .Conclusion :Intracerebral hemorrhage can be induced in mouse by injecting collagenase type Ⅳ into the corpus striatum of the brain .This is a useful economical and reproducible method to make intracerebral hemorrhage .ICH model was made by intracerebral injection of 0 .02U type IV collagenase can meet our experimental requirements .
