摘要 目的建立加速溶剂萃取-高效液相色谱-电喷雾-离子阱质谱(ASE-HPLC-ESI-IT/MS)联用技术分析淡豆豉中大豆异黄酮类化合物的方法。方法采用加速溶剂萃取仪,以70%(体积分数)乙醇为溶剂提取淡豆豉中的大豆异黄酮类成分,色谱柱为XAqua C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%(体积分数)甲酸,梯度洗脱。离子阱质谱分别采用正、负离子模式下扫描,对提取出的大豆异黄酮类成分进行鉴定。结果从淡豆豉中共提取鉴别出9种结合型糖苷类大豆异黄酮化合物。结论该方法优势显著,操作简便、灵敏、精确,适宜于淡豆豉中大豆异黄酮的分析鉴定。 Objective To establish an accelerated solvent extraction-high performance liquid chromatography- eleetrospray ionization-ion trap-mass spectrometry (ASE-HPLC-ESI-IT/MS) analysis method for the analysis of soybean isoflavones in Semen sojae praeparatum. Methods The samples were extracted by ASE and the extraction solvent was 70% ethanol.The chromatographic analysis was performed on a XAqua C18 column (250 min×4.6 ram,5 p,m) by a gradient elution of acetonitrile and 0.1% formic acid as mobile phases. The soybean isoflavones were detected and identified by electrospray ionization-ion trap-mass spectrometry both in positive and negative ion modes. Results 9 soybean isoflavones were extracted and identified successfully by this method.Conclusion The method is simple, sensitive, accurate with significant advantages, and suitable for the analysis and identification of the soybean isoflavones in Semen sojae praeparatum.
