摘要
目的建立消渴丸中黄芪甲苷的含量测定方法。方法用高效液相色谱法,以蒸发光散射检测器测定黄芪甲苷的含量。色谱柱为Thermo—C18(4.6mm×250mm,5um),流动相为乙腈-水(32:68),流速为:1.0mL·min-1;ELSD参数气体流速为1.60mL·min-1,漂移管温度为80℃;进样量为20uL。结果黄芪甲苷进样量在1.016~4.064ug范围内呈现良好的线性关系,r:0.9998(n=5);平均回收率为97.03%,RSD=1.8%(n=9)。结论所建立的含量测定方法操作简便、快速、准确、重现性好,可作为消渴丸的质量控制。
Objective To establish a method for the determination of astragaloside in Xiaoke Pills. Methods The HPLC - ELSD was conducted on a Thermo - ClS column (4.6 mm × 250 mm,5 um ) with acetonitrile - water ( 32:68 ) as mobile phase, the flow rate was 1.0 mL · min - 1. The gas flow rate of ELSD parameter was 1.60 mL · min - 1. The drift tube temperature was 80℃ ,the injection volume was 20 uL. Results The linear range of astragaloside was 1. 016 - 4. 064 ug, r = 0. 999 8 ( n = 5 ) , the recovery rate was 97.03 % , RSD = 1.8 % ( n = 9 ). Conclusion The established HPLC - ELSD was simple ,rapid ,accurate ,reproducible ,and can be used for the quality control of Xiaoke Pills.
