软肝化坚颗粒对肝纤维化C57小鼠体内羟脯氨酸的影响 被引量:3

摘要 目的观察软肝化坚颗粒对肝纤维化模型C57小鼠肝组织和血清中羟脯氨酸(Hyp)的影响。方法将40只C57小鼠随机分为4组,即正常对照组、模型对照组、模型软肝化坚颗粒组及模型秋水仙碱组,每组10只。采用四氯化碳腹腔注射制作小鼠肝纤维化模型,造模同时模型软肝化坚颗粒组采用软肝化坚颗粒进行治疗,模型秋水仙碱组采用秋水仙碱进行治疗,用药6周后,分离小鼠外周血血清用酶联免疫吸附法(ELISA法)检测Hyp、转化生长因子β1(TGF-β1)和Ⅰ型胶原的含量,采集各组小鼠肝组织样本,碱水解法检测肝组织中Hyp的含量。结果正常对照组、模型软肝化坚颗粒组和模型秋水仙碱组小鼠血清Hyp、TGF-β1和Ⅰ型胶原的含量及肝组织中Hyp含量均低于模型对照组(P<0.05)。结论软肝化坚颗粒可以有效降低小鼠肝组织和血清中Hyp的含量,这可能是软肝化坚颗粒防治肝纤维化的机制之一。 Objective To study the effect of Ruangan Huajian granule on Hyp Level in serum and hepatic tissue of C57 mice with experimental hepatic fibrosis .Methods Forty C57 mice were randomly divided into four groups,i.e.normal control group,model control group,Ruangan Huajian granule model group ,and colchicine model group.There were 10 mice in each group.Experimental mice with hepatic fibrosis were induced by CCl 4 intraperitone-ally.Mice in colchicine model group were treated by colchicine .Mice in Ruangan Huajian granule model group were treated by Ruangan Huajian granule .The levels of serum Hyp ,TGF-β1 and typeⅠcollagen were detected by ELISA in the four groups .And the levels of hepatic tissue Hyp were detected in the four groups .Results The levels of ser-um Hyp,TGF-β1 and type I collagen in Ruangan Huajian Particle model group ,colchicine model group and normal control group were notably lower than those in model control group (P〈0.05).The levels of hepatic tissue Hyp in Ruangan Huajian Particle model group ,colchicine model group and normal control group were notably lower than those in model control group (P〈0.05).Conclusion Ruangan Huajian granule can decrease Hyp level in serum and he-patic tissue of C57 rats with experimental hepatic fibrosis .It has to some extent preventive and therapeutic value for experimental hepatic fibrosis in rat models .
