大针手法针刺督脉对癫痫大鼠海马神经细胞核转录因子-κB表达的影响 被引量:12

Effects of Nuclear Factor-kappa B Expression in Hippocampal Nerve Cells of Epileptic Rats through Dazhen Acupuncture Manipulation on Du Meridian

摘要 目的研究大针手法针刺督脉对癫痫大鼠海马神经细胞核转录因子-κB(nuclear factor-kappaB,NF-κB)表达的影响,探讨大针手法针刺督脉治疗癫痫的作用机制。方法 30只健康Wistar大鼠,体重200g^250g,雌雄各半,随机分为空白对照组(A组)、癫痫模型组(B组)、大针手法组(C组),每组10只,雌雄各半,癫痫模型组和大针手法组大鼠腹腔注射氯化锂(3mmol/kg)和毛果芸香碱(匹鲁卡品,30mg/kg,分3次注射),诱导癫痫持续状态(status epilepticus,SE)大鼠模型,观察大鼠的行为学改变,空白对照组腹腔注射同等剂量的生理盐水代替氯化锂和匹鲁卡品。空白对照组和癫痫模型组不做任何治疗,大针手法组用1寸不锈钢针,针刺大鼠督脉穴:风府、大椎、陶道、无名、身柱,每日1次,每次20min,中间行针一次,平补平泻法,治疗14d。运用HE染色法观察3组大鼠海马神经细胞病理形态学改变,运用免疫组化(SABC)法观测3组大鼠海马神经NF-κB P65阳性细胞数的表达。结果癫痫模型组和大针手法组大鼠均诱导出癫痫持续状态,两组各死亡1只。HE染色显示,癫痫模型组大鼠海马神经细胞出现明显的病理改变,大针手法组大鼠海马神经细胞病理改变明显改善,空白对照组大鼠神经细胞未见明显病理改变。免疫组化染色:3组大鼠海马神经NF-κB P65阳性细胞数的表达依次为(13.95±6.99)个、(28.75±8.80)个、(19.25±8.60)个,癫痫模型组、大针手法组与空白对照组比较,差异有统计学意义(P<0.05),大针手法组与癫痫模型组比较,差异有统计学意义(P<0.05)。结论大针针刺督脉可能是通过抑制癫痫大鼠海马神经细胞NF-κB的表达,从而减少海马神经细胞的凋亡,达到治疗癫痫的目的。 Objective To study the effects of nuclear factor - kappa B(NF - κβ) expression in hippocampal nerve cells of epileptic rats through dazhen acupuncture manipulation on Du Meridian, then explore the mechanism of anti-epilepsy through dazhen acupuncture manipulation on Du Meridian, provide new theoretical basis for clinical acupuncture treatment of epilepsy. Methods Thirty health rats were randomly and enenly divided into blank control group(group A), epilepsy model group(group B), dazhen acupuncture manipulation group(group C), Lithium chloride and pilocarpine were injected the rats' enterocoelias of group B and group C,induce status epilepticus(SE) model. The rats' behavior changes were observed. Equal dose of normal saline were injected the rats' enterocoelia of group A. Any treatment was not given to group A,dazhen acupuncture manipulation was applied at points of Du Meridian : Fengfu, Dazhui, Taodao, Wuming, Shenzhu, once a day, 20 minutes every time, treatment for 14 days. The pathological morphological changes in the hippocampal nerve cells of the three groups were observed by using HE staining. The expression of NF - κβ P65 in hippocampal nerve cells of the epileptic rats were observed by immunohistoehemistry. Results The status epilepticus were induced in group B and group C,one rat dead in two groups. The rats in group B had obvious pathological morphological changes,the pathological morphological change in group C had obvious improvement. There was no obvious pathological morphological changes in group A. Immunohistochemical staining : The expression of NF - κβ P65 was( 13.95 ± 6.99) in group A, (28.75 ± 8.80) in group B, (19. 25±8.60) in group C. Comparing each two groups,the differences are statistically significant(P〈0.05). Conclusion Dazhen acupuncture manipulation on Du meridian, maybe can suppress the expression of NF- κB P65 in the hippocampal nerve cells of epileptic rats, then reduce the hippocampal nerve cells apoptosis,treat epilepsy.
