蛋白载体温敏凝胶制备及释放度研究 被引量:3

Preparation of Bovine Serum Albumin in Thermal Sensitive Gel and Determination of Its Release Percent

摘要 为研究温敏凝胶作为疫苗或多肽蛋白类药物的佐剂或载体的可行性,本研究采用泊洛沙姆为温敏凝胶基质,以牛血清白蛋白为模型蛋白配制了两种不同配比的凝胶,采用试管倒置法,分别测定了添加牛血清白蛋白前后的胶凝温度的变化,采用Bicinchoninic acid蛋白含量测定法测定了以水为介质的BSA释放度,结果显示,添加BSA后胶凝温度和空白凝胶无差异,且两种不同配比的释放行为无显著差异,均为一级释放动力学,且释放机制为Fickian扩散。 To study the feasibility of temperature-sensitive gel used as a vaccine adjuvant or protein drugs carrier, this study used Poloxamer gel as temperature sensitive gel and bovine serum albumin as a model protein to prepare two formulations with different ratios of gel. The gelling temperatures both of blank gel and BSA gel were detected by an inverted test tube method. And the release contents of BSA gels in water were detected BCA kit. The results showed that there was no significant difference of the gelling temperatures or release percents between blank gel and BSA gel. The release processes of both of two ratio gels were conformed to first order equation and the release mechanisms were Fickian diffusion.
