健胎液对胚胎着床障碍小鼠子宫炎性细胞因子表达的影响

Effects of Jiantai Liquid on the Expressions of Uterine Inflammatory Cytokines in Mice With Embryo Implantation Dysfunction

目的探讨中药健胎液对子宫内膜着床微环境改善的可能分子机制。方法 90只受孕小鼠随机分为正常组、模型组、中药组各30只,模型组和中药组采用吲哚美辛皮下注射建立胚胎着床障碍小鼠模型。中药组从妊娠第1天开始用健胎液(144%无菌煎剂)灌胃,每天每只0.5ml;模型组和正常组每天用等量生理盐水灌胃。在妊娠第5天检测各组小鼠(每组10只)子宫内膜白介素-1β(IL-1β)、白介素1受体Ⅰ型(IL-1RⅠ)、白介素-6(IL-6)、白血病抑制因子(LIF)、集落刺激因子-1(CSF-1)mRNA水平,妊娠第8天检查各组小鼠(每组20只)子宫内胚胎发育情况,记录胚胎着床率和着床位点数。结果中药组和模型组的胚胎着床率、着床位点数同正常组比较显著降低(P<0.01),中药组胚胎着床率、着床位点数同模型组比较明显升高(P<0.05)。与正常组比较,模型组IL-1β、IL-1RⅠ、IL-6、LIF、CSF-1 mRNA含量均显著升高(P<0.01);与模型组比较,中药组IL-1β、IL-1RⅠ、IL-6、LIF、CSF-1 mRNA含量均显著降低(P<0.05或P<0.01)。结论中药健胎液通过降低胚胎着床障碍小鼠子宫内膜IL-1β、IL-1RⅠ、IL-6、LIF、CSF-1细胞因子的基因表达,改善子宫内膜微环境,促进着床。

Objective To study the possible molecular mechanism of Jiantai Liquid in improving the implantation environment in the endometrium. Methods Totally 90 pregnant mice were randomized into the normal group, model group and Jiantai Liquid group, with 30 in each. The embryo implantation dysfunctional （EID） models were established with indomethacin in the model group and Jiantai Liquid group. The Jiantai Liquid group was gavaged with Jiantai Liquid from the 1 st day of pregnancy. The model group and normal group were gavaged with the same amount of normal saline. The levels of interleukin-1β （IL-1β）, IL-1 receptor type I （IL-1RI）, IL-6, leukemia inhibitory factor （LIF） and colony stimulating factor-1 （ CSF-1 ） mRNA were detected on the 5th day of pregnancy. The embryonic development was observed, and the numbers and site numbers of embryo implantation were recorded on the 8th day of pregnancy. Results Comparing with the normal group, the numbers and site numbers of embryo implantation were significantly decreased in the Jiantai Liquid group and model group （ P 〈 0. 01 ）. Comparing with the model group, the numbers and site numbers of embryo implantation were significantly increased in the Jiantai Liquid group （P 〈 0. 05）. Comparing with the normal group, the contents of IL-1β, IL-1RI, IL-6, LIF and CSF-1 mRNA were significantly increased in the model group （P 〈 0. 01 ）. Comparing with the model group, the contents of IL-1β, IL-1RI, IL-6, LIF and CSF-1 mRNA were significantly decreased in the Jiantai Liquid group （ P 〈 0. 01 ）. Conclusion Jiantai Liquid can improve endometrial microenvironment and promote implantation by deregulating the expressions of IL-1β, IL-1RI, IL-6, LIF and CSF-1 mRNA
