Wnt信号通路抑制因子DKK-1蛋白的实验研究进展 被引量:3

摘要 DKK-1(Dickkopf-1)蛋白作为Wnt信号通路的抑制因子之一,可通过竞争性结合脂蛋白受体相关蛋白5/6(LRP 5/6),也可通过与跨膜蛋白kremen、LRP 5/6形成三元复合物而导致快速内吞,减少质膜LRP 5/6而抑制Wnt信号通路。近期研究发现DKK-1在抗肿瘤、抗组织器官纤维化、调节骨代谢、预测心脑血管事件发生等方面发挥重要作用,从而为临床诊断治疗提供了新的思路。本文旨对DKK-1的实验研究进展进行综述。 DKK-1 (Dickkopf-1),as one of the inhibitors of Wnt signaling pathway,block Wnt signaling pathway by competitive binding lipoprotein-receptor-related proteins 5 and 6 (LRP 5/6) or forming a ternary complex with kremen and LRP 5/6, which induces rapid endocytosis and removal of the Wnt receptor LRP 5/6 from the plasma membrane. Recently, research shows that DKK-1 play an important role in anti-tumor therapy, anti fibrosis of tissue and organ therapy, regulating bone metabolism and predicting the risk of cardiovascular events etc, which provide a new idea for clinical diagnosis and treatment. The paper will review advances in the experimental research of DKK-1.
