摘要
[目的]建立4个多用途萱草品种的离体快繁体系。[方法]以4个多用途萱草品种为试材,研究消毒时间对不同外植体的影响,以及不同激素配比的培养基对愈伤组织、增殖培养和生根培养的影响。[结果]消毒时间以10 min最佳,花茎是萱草组织培养的最佳外植体;愈伤组织诱导的最佳培养基为:MS+1.5 mg/L 6-BA+0.2 mg/L IBA;在增殖培养中,MS+1.5 mg/L 6-BA+0.2 mg/L IBA为最佳增殖培养基。1/2MS+0.3 mg/L IBA和1/2MS+0.3 mg/L NAA培养基为4个品种的最佳生根培养基。[结论]该方法建立了4个多用途萱草品种的离体快繁体系,为萱草的种质资源栽培提供了理论依据。
[Objective] To establish tissue culture and rapid propagation of the four multi-role Hemerocallis fulva.[Method] With the four multi-role H.fulva as test materials,effects of disinfection time on different explants were studied,effects of culture medium with different hormone ration on tissue callus,proliferation and rooting culture were studied.[Result] The results showed that 10 min was the best disinfection time,and the best explant was scape; The best medium for callus was MS + 1.5 mg/L6-BA + 0.2 mg/L IBA ; Shoot reproduction medium was MS + 1.5 mg/L6-BA + 0.2 mg/L IBA ; The best rooting medium was 1/2MS + 0.3 mg/L IBA and 1/2MS + 0.3 mg/L NAA.[Conclusion] The tissue cuhure and rapid propagation of four multi-role Hemerocallis fulva were established,which will provide theoretical basis for germplasm resource cultivation of Hemerocallis fulva.
