培养基组成对布拉氏酵母生长的影响及其优化 被引量:5

摘要 对益生菌布拉氏酵母(Saccharomyces boulardii)的培养基组成及培养条件对其生物量的影响进行了研究。通过单因素试验探讨培养基中碳氮源种类,磷酸二氢钾和硫酸镁浓度、pH以及接种量对布拉氏酵母生物量的影响;利用正交试验确定摇瓶培养的最适条件。结果表明布拉氏酵母的发酵培养基组成为:葡萄糖45 g/L,酵母膏12 g/L,磷酸二氢钾0.8 g/L,硫酸镁0.5 g/L。培养条件为:培养基初始pH5.5,接种量9%,培养时间24 h。在此条件下得到的布拉氏酵母菌体干重为6.5 g/L。 Effects of fermentation medium components and conditions on growth of Saccharomyces boulardii were investigated in this study separately, based on the single-factor experiment and the orthogonal test. The influential factors such as carbon and nitrogen sources, KH2PO4 and MgSO4 concentrations, pH and incubation size were studied respectively. Results of the orthogonal test demonstrated that S.boulardii grew better in medium (per liter) consisted of 45 g of glucose, 12 g of yeast extract, 0.8 g of KH2PO4, 0.5 g of MgSO4, and the optimal conditions for growth of S.boulardii required initial pH of 5.5, incubation size of 9%and fermentation time of 24 h. Under these conditions, the dry weight of S.boulardii reached to 6.5 g/L.
