摘要
研究旨在利用C57小鼠60Coγ射线辐照模型,观察睾酮衍生物ATES对小鼠照射后外周血血象的影响。48只C57小鼠经6.5 Gy60Coγ射线全身1次照射后建立骨髓型急性放射病动物模型。模型动物被随机分为照射对照组、单次给药组、照后连续n次给药组(n=2、3、4、5),每组8只。单次给药组于照射后2 h皮下注射ATES 10 mg/kg;连续给药组分别于照射后2 h和照后n d内每天一次皮下注射ATES 10 mg/kg;照射对照组于皮下注射同样体积的空白辅剂。观察照射前后小鼠外周血象变化与药物对体重的影响。与照射对照组相比较,ATES治疗能明显促进外周血白细胞数、中性粒细胞数、红细胞数、血红蛋白水平、血小板数以及单核细胞数的恢复。结论:ATES可以明显促进急性放射病小鼠的造血功能恢复,为研究临床治疗急性放射病提供实验依据。
A C57 mice model of 60Co γ–ray irradiation was observed Testosterone derivatives ATES on mice peripheral blood after irradiation effects. The bone marrow failure caused by severe acute radiation exposure model was established in 48 C57 mice which received total body irradiation by 6.5 Gy 60Co γ in single time irradiation. The irradiated mouse were randomly divided into 6 subgroups: controls (n=8) which received subcutaneous blank adjuvants, ATES single-dose group (n=8) which received subcutaneous injection of ATES (10 mg /kg) at 2h after irradiation, two-dose group (n=8)which received subcutaneous injection of ATES (10 mg /kg) at 2h ,day1 after irradiation, three-dose group (n=8) which received subcutaneous injection of ATES (10 mg /kg) at 2h ,day1 and 2 after irradiation, four-dose group (n=8) which received subcutaneous injection of ATES (10 mg /kg) at 2h, day1, 2 and 3 after irradiation, five-dose group (n=8) which received subcutaneous injection of ATES (10 mg /kg) at 2h ,day1, 2, 3 and 4 after irradiation respectively. Peripheral blood cells and weight were counted before and irradiation. Compared with control group, ATES administrated mouse’ white blood cell count, neutrophile granulocyte, red blood cell count, hemoglobin level, platelet, monocyte showed fasted recovery from high dose radiation. It is concluded that ATES treatment can significantly promote mice which suffered from bone marrow failure caused by severe acute radiation exposure bone marrow hematopoietic function recovery, to study clinical treatment of acute radiation sickness provide experiment evidence.
