摘要
目的 探讨热休克蛋白70(HPS70)在兔非酒精性脂肪肝(NAFLD)纤维化形成过程中的作用.方法 40只雄性日本大耳兔随机分为正常对照组和NAFLD模型组,其中对照组取8周为时相点,模型组取4周、6周、8周3个时相点,每个时相点各10只.模型组分别皮下注射花生油4周、6周、8周,2次/周,每次1.2 ml/kg,建立日本大耳兔NAFLD模型.按时间点处死各组动物,测定兔血清中ALT、AST、TC、TG、FFA、空腹血糖(FBG)、HSP47、HSP70、HA、PC Ⅲ、C Ⅳ、LN的含量和空腹胰岛素(FINS)的水平,并计算胰岛素抵抗指数(HOMA-IR); MASSON染色观察肝组织病理形态改变;免疫组化观察HSP47和HSP70的表达情况.结果 与对照组相比,模型组肝功能、脂质代谢和肝纤维化相关指标水平升高,造模4周时出现肝功能损伤,脂质代谢紊乱、胰岛素抵抗,随着NAFLD肝纤维化进程的发展,HSP70表达量逐渐增加.结论 HSP70的表达与NAFLD肝纤维化程度呈正相关,可能与NAFLD肝纤维化发生发展密切相关.
Objective To investigate the change of HSP70 in nonalcoholic fatty (NAFLD) liver fibrosis of rabbit. Methods Forty male Japanese white rabbits were randomly assigned to control group and model group. The model group was divided into three subgroups, as 4-week, 6-week, 8-week groups, with 10 rabbits in each, and treated with peanut oil (1.2 ml/kg) subcutaneously (s.c.), twice a week for 4 weeks, 6 weeks and 8 weeks respectively. The rabbits of all groups were sacrificed at the designed time point. Samples of serum were collected for detecting the serum ALT, AST, TC, TG, FFA, FBG, HSP47, HSP70, HA, PC?, C/∞, LN ,and the HOMA-IR were calculated. The liver tissues were collected for histopathological and immunohistochemical observation. Results The model groups had hepatic injury and high level of insulin at the beginning of 4 weeks. With the process of NAFLD liver fibrosis, HSP70 had gradually increased. Conclusion The expression of HSP70 was positively corre- lated with NAFLD liver fibrosis, and it might have close relation with the development of NAFLD liver fibrosis.
