Neutrophil extracellular traps promote thrombus formation through interaction with platelets

摘要 人类许多严重疾病,如脓毒症、创伤、缺血、癌症和自身免疫性疾病与血栓形成有关。中性粒细胞和血小板共同位于血管损伤、出血和血栓形成部位,在炎症、止血和血栓形成过程中发挥重要作用。目前认为,中性粒细胞胞外诱捕网(neutrophil extracellular traps,NETs)或细胞外组蛋白与血小板的相互作用促进血栓形成,为临床预防和治疗血栓性疾病提供了新的研究领域。 Neutrophils release neutrophil extracellular traps(NETs)in response to microbial and inflammatory stimuli.NETs cause platelet adhesion,activation and aggregation,and provide a stimulus and scaffold for thrombus formation.Histones in NETs or liberated after digestion of NETs can also provide a stimulus for platelet aggregation.Extracellular histones promote thrombin generation through Toll-like receptors(TLR2/TLR4)on platelets and platelet-derived inorganic polyphosphate(polyp).NETs/histones are a unique link between inflammation and thrombosis and may further explain the epidemiological association of infection with thrombosis.
