苍附导痰丸加减对多囊卵巢综合征大鼠肿瘤坏死因子受体的影响 被引量:1

摘要 目的探讨苍附导痰丸加减(TCM-CFDTW)对多囊卵巢综合征(PCOS)大鼠肿瘤坏死因子受体(TNFR1、TNFR2)表达的影响,明确TCM-CFDTW治疗PCOS的机制。方法取雌性大鼠随机分成5组,每组大鼠10只。模型组采用丙酸睾酮+绒毛膜促性腺激素(HCG)建立PCOS大鼠病理模型;克罗米芬(CC)组造模结束后,第1-14天灌胃给予CC悬液;中药高剂量组造模结束后给予TCM-CFDTW 24 g/kg灌胃;中药低剂量组造模结束后24 h用TCM-CFDTW 6 g/kg灌胃;空白对照组未造膜,大鼠颈背部皮下注射等量的生理盐水并灌胃等量生理盐水。14 d后光镜观察卵巢大体形态变化、卵泡发育情况,放射免疫法测定血清雄激素(T)、雌二醇(E2)、促黄体生成素(LH)、卵泡刺激素(FSH),免疫组织化学法测定卵巢组织TNFR1、TNFR2表达。结果中药高浓度组及CC组均能显著改善大鼠的激素水平(P〈0.05),以中药高浓度组最为明显;中药高浓度组能显著减少TNFR1、TNFR2蛋白表达(P〈0.05),CC组能减少TNFR2蛋白表达(P〈0.05)。结论 TCM-CFDTW对丙酸睾酮+HCG建立的PCOS大鼠有一定的治疗作用,其机制可能是通过调节TNFR1、TNFR2蛋白表达,从而抑制了炎症信号的通路。 Objective to investigate the cfdt pill( TCM- CFDTW) on polycystic ovary syndrome( PCOS) rat tumor necrosis factor receptors( TNFR1,TNFR2) on the expression of TCM- CFDTW in the treatment of PCOS mechanism, clear. Methods the female rats were randomly divided into 5 groups,each group 10 rats. Model group with testosterone propionate + human chorionic gonadotropin( HCG) to establish PCOS rat animal model; clomiphene citrate( CC) group after model establishment,the first 1 - 14 days by intragastric administration of CC suspension; traditional Chinese medicine high dose group after giving TCM-CFDTW 24 g / kg gavage; low dosage Chinese medicine group after modeling 24 h TCM- CFDTW 6 g / kg gavage; saline control group did not cause membrane of rat subcutaneous injection of equivalent and to intragastric normal saline. 14 d gross morphological changes were observed under optical microscope,ovarian follicular development,the determination of serum testosterone radioimmunoassay( T),estradiol( E2),luteinizing hormone( LH),follicle stimulating hormone( FSH),TNFR1,TNFR2 expression of ovarian tissue by immunohistochemistry. Results The high concentration of traditional Chinese medicine group and CC group could significantly improve the hormone level in rats( P〈0. 05),with high concentration of traditional Chinese medicine traditional Chinese medicine was most significant; high concentration group can significantly reduce the expression of TNFR1,TNFR2 protein( P〈0. 05),CC group can decrease the expression of TNFR2 protein( P〈 0. 05). Have a certain therapeutic effect on PCOS rats Conclusion TCM-CFDTW testosterone propionate + HCG is established,and its mechanism may be through regulating TNFR1 and TNFR2 protein expression,thus inhibiting the inflammatory signal pathway.
