摘要
目的明确ZNT1在膀胱癌组织中表达情况,为进一步的研究奠定基础。方法免疫组化检测23例膀胱癌和9例正常膀胱组织中ZNT1的表达情况;利用Real-time RT-PCR和Western blot法检测膀胱组织中ZNT1的mRNA和蛋白表达情况,检测ZNT1在膀胱癌组织中mRNA和蛋白的表达情况,分析ZNT1基因表达的临床意义。结果免疫组化检测结果显示ZNT1在正常膀胱组织中表达非常微弱,其阳性率为22.2%,而在膀胱癌组织中其阳性率为87%,差异有统计学意义(P<0.01)。Real Time RT-PCR及Western Blot结果均显示ZNT1在膀胱癌组织中表达显著升高,与其在正常膀胱组织中的表达量相比,差异有统计学意义(P<0.01)。结论 ZNT1在mRNA及蛋白水平,在膀胱癌中的表达含量与其在正常膀胱组织的表达量相比均显著升高,有可能成为膀胱癌的一个早期诊断指标。
Objective To detect the expression levels of zinc transporter (ZNT1) in normal bladder mucosal tissue and bladder carcinoma tissues in order to provide scientific data for further study. Methods Immunohistochemistry was used to detect the expression levels of ZNT1 in 23 cases of bladder cancer tissue and 9 cases of normal bladder tissue. Real Time RT-PCR and Western Blot were used to detect the mRNA and protein levels of ZNT1 in bladder carcinoma and normal bladder mucosal tissue so as to analyze the clinical significance of ZNT1 gene expression. Results The immunohistochemical results showed that the expression levels of ZNT1 in normal bladder tissue were relatively lower, with the expression positive rate being 22.2% ,however,which in bladder cancer tissue was 87% , there were significant differences between normal bladder tissue and bladder cancer tissue ( P 〈 0.01 ). Both the Real Time RT-PCR and Western Blot analysis results showed that the expression levels of ZNT1 in normal bladder tissue were significantly increased, as compared with those in normal bladder tissues ( P 〈 0.01 ). Conclusion The expression levels of ZNT1 mRNA and protein in bladder cancer tissue are obviously increased,as compared with those in normal bladder tissue,which may be one of indexes in the early diagnosis of bladder cancer.
