摘要
目的探讨survivin、caspase-3在乳腺癌细胞中的表达及与乳腺癌细胞凋亡及增殖的相关性。方法采用免疫组织化学聚合酶两步法(PV法)检测乳腺癌、乳腺腺瘤及乳腺正常组织中survivin、caspase-3及PCNA的表达情况,TUNEL试剂盒检测细胞凋亡情况。结果乳腺癌组织中survivin、caspase-3表达的OD值、增殖指数和凋亡指数与乳腺癌及乳腺正常组织比较,差异有统计学意义(P<0.05)。病理分级越低、外科分期越高,survivin表达的OD值及AI越高,caspase-3表达的OD值及AI越低(P<0.05)。survivin蛋白的表达与乳腺癌细胞凋亡指数(AI)呈负相关(r=-0.707,P<0.05),而与增殖指数(PI)正相关(r=0.302,P<0.05);caspase-3蛋白的表达与乳腺癌细胞PI呈负相关(r=-0.028,P<0.05),而与AI正相关(r=0.319,P<0.05);survivin的表达与caspase-3呈负相关(P<0.05)。结论乳腺癌的发生发展中可能普遍存在细胞凋亡、增值机制的紊乱,survivin,caspase-3的异常表达在乳腺癌的发生发展中起重要作用。
Objective To investigate the expression of survivin and caspase -3 in breast cancer cells,and to explore its correlation with apoptosis and proliferation of breast cancer .Methods Immunohistochemical method (PV) was used to detect the expressions of survivin,caspase-3 and PCNA in 300 cases of breast cancer,132 cases of breast adenoma and 103 cases of breast normal tissue.TUNEL reagent box was used to examine apoptosis cell .Results There were significant differences in the OD value of surviving,caspase-3 expression,proliferation index (PI) and apoptosis index (AI) between breast cancer tissues and breast adenoma ,breast normal tissue ( P 〈0.05).The lower pathological classification and the higher surgery staging of breast cancer were ,the higher OD value of surviving expression and AI ,however,the lower OD value of caspase-3 expression and AI ( P 〈0.05).The expression of surviving was negatively correlated to AI of breast cancer cells ( r =-0.707, P 〈0.05),however,which was positively correlated to PI ( r =0.302, P 〈0.05).The expression of caspase-3 was negatively correlated to PI of breast cancer cells ( r =-0.028, P 〈0.05),however,which was positively correlated to AI ( r =0.319, P 〈0.05).The expression of surviving was negatively related with that of caspase -3 ( P 〈0.05).Conclusion The cell apoptosis and disorder of cell proliferation mechanism exist generally in the pathogenesis and development of breast cancer .The abnormal expressions of survivin and caspase -3 may play an important role in the pathogenesis and development of breast cancer .
