摘要 为探究褐色菜鸭POU1F1基因的多态性及其遗传效应。本试验采用PCR-SSCP方法检测了褐色菜鸭POU1F1基因的多态性,分析了多态位点不同基因型与褐色菜鸭生产性状的关联性。结果表明:POU1F1基因第2内含子存在3种基因型即AA型、Aa型和aa型,序列比对存在2个突变位点即A3980T和A3988G,该位点属低度多态,c2检验结果表明:该基因座位处于Hardy-Weinberg平衡(P>0.05);关联分析显示:AA基因型个体的蛋形指数和四周龄体重均显著高于aa型(P<0.05),但三种基因型对其他性状无显著影响(P>0.05)。推测该片段的多态位点可能对四周龄体重和蛋形指数有一定的影响,POU1F1基因第2内含子可作为褐色菜鸭生产性能的优势分子标记。 This study aimed to detect the polymorphism of POU1F1 gene and its genetic effects on Brown Tsaiya Duck. PCR-SSCP method was applied to detect the polymorphisms of POU1F1 gene, and analyze the association of polymorphisms with production traits of Brown Tsaiya Duck. The results showed that POU1F1 intron2 was consisted of 3 genotypes(AA, Aa, aa). 2 SNPs loci(A3980T, A3988G) were found. This site belongs to low polymorphism, and the result of c2indicated that the site was in state of Hardy-Weinberg equilibrium. Both egg shape index and body weight at 4th week of AA genotype were significantly higher than that of aa(P〈0.05), while three kinds of genotypes had no significant effect on other indexes(P〉0.05). The results suggested that the SNPs had certain effect on egg shape index and body weight at 4thweek and might be a good marker for production traits in Brown Tsaiya Duck.
