大别山石斛外植体的选择与组织培养技术研究

Research on the Explants Selection and Tissue Culture Technology of Caulis Dendrobe from Dabie Mountain

以大别山石斛[Dendrobium dabieshanense(C.Z.Tang et S.J.Cheng)]为材料,探讨了不同外植体、培养基中不同的生长素配比、不同成分的大量元素及部分微量元素含量对不定芽分化和试管苗生根的影响。结果表明,适宜大别山石斛组培的外植体是种子,其次是茎;不定芽增殖的适宜培养基是1/2MS+马铃薯汁10g/L+IBA 0.1mg/L+琼脂8g/L+食糖30g/L,pH5.6;试管苗生根的适宜培养基是1/2MS+香蕉汁5g/L+IBA 0.2mg/L+琼脂8g/L+食糖30g/L,pH5.6。

Choosing the dendrobe[Dendrobium dabieshanense(C.Z.Tang et S.J.Cheng)]from Dabie Mountain as the experimental material,the effect on the adventitious bud differentiation and rooting of plantlet in test tube of different explants,different medium contains different match of various auxin,various major elements and contents of some microelements by the above different media were studied.The results indicated that the seed was more suitable explant of Dabie Mountain dendrobe,and the secondly suitable explant was the stem;the suitable medium for adventitious bud proliferation was 1/2MS+potato juice 10g/L +IBA 0.1mg/L+agar 8g/L+sugar 30g/L,pH5.6;the suitable medium for rooting of plantlet in test tube was 1/2MS +banana juice 5g/L+IBA 0.1mg/L+agar 8g/L+sugar 30g/L,pH5.6.
