类泛素分子FAT10上调核因子Kappa B表达与糖尿病肾病关系的研究进展

摘要 糖尿病肾病(diabetic nephropathy,DN)是糖尿病主要的微血管并发症,也是糖尿病主要死亡原因之一.核因子Kappa B(NF-κB)是一种广泛存在于真核细胞内的转录因子,可通过调节转化生长因子β1(TGF-β1)表达、促进细胞凋亡等途径在进展期DN病程中占据重要地位.人类白细胞抗原F介导转录因子10(human leukocyte antigen F-associated transcript 10,FAT 10)是类泛素分子家族(ULMs)成员,具有与底物蛋白共价结合,并介导底物蛋白快速而不可逆地被蛋白酶体靶向降解的特性.研究表明,FAT10在受细胞因子等刺激后,可促进NF-κB通路抑制蛋白IκB-α迅速降解,上调NF-κB表达,而后者可以进一步促进DN发展.本文将就FAT10、NF-κB及DN三者之间关系的研究进展作一综述.
