摘要
目的探讨类风湿关节炎(RA)关节液和滑膜组织中人诱骗受体3(DcR3)表达水平变化,以及关节滑膜组织炎性细胞数与DcR3表达水平的相关性。方法采用免疫细胞化学法,扫描分析了30例RA患者、20例骨关节炎(OA)患者和30例关节正常的骨折患者滑膜组织中DcR3阳性细胞,并计数其中的炎性细胞;用酶联免疫吸附试验测定30例RA患者和20例OA患者关节液中DcR3浓度,将试验数据进行统计学处理。结果 RA患者DcR3在滑膜细胞中高表达,阳性率约72%,并且与炎性细胞数呈正相关(r=0.989,P<0.01);RA患者关节液DcR3浓度显著高于OA患者,差异具有统计学意义(P<0.01)。结论 RA患者滑膜组织和关节液中DcR3高表达可能与自身免疫性炎症反应有关,DcR3可能参与类风湿关节炎的发病机制。
Objective To discuss the expression level of human decoy receptor 3(DcR3) in synovial fluid and synovial tissue of patients with rheumatoid arthritis( RA),and investigate the relationship between the number of inflammatory cells and DcR3. Methods By using immunocytochemical method,30 RA patients,20 osteoarthritis patients( OA) and 30 fracture patients with normal joints were involved to analyze the number of DcR3 positive cells in the synovial tissue,and count the amount of inflammatory cells. Enzyme- linked immunosorbent assay was used to determine the DcR3 density in the joint fluid of 30 RA patients and 20 OA patients,then the determination results were analyzed by statistics. Results DcR3 was highly expressed in synovial cells of RA patients,the positive rate was about 72%,and significantly positively correlated with the number of inflammatory cells(r = 0. 989,P〈0. 01); DcR3 concentration in synovial fluid of RA patients were higher than that in OA patients, and the difference had statistical significance(P〈0. 01). Conclusion High DcR3 expression in synovial tissue and synovial fluid may be related with autoimmune inflammation in RA patients,indicating that DcR3 may be involved in the pathogenesis of rheumatoid arthritis.
