雷公藤生物碱系列组分对人CIK、HCT116及HepG2细胞的增殖抑制作用 被引量:6

摘要 目的从雷公藤生物碱系列组分中筛选具有免疫抑制活性及抗肿瘤作用的组分,评价其增效减毒效果。方法采用MTT法,研究雷公藤总生物碱及倍半萜类生物碱共5种组分对人CIK细胞、结直肠腺癌HCT116细胞及肝癌HepG2细胞的增殖抑制作用,以及筛选出的LGT-ZF1组分在不同时间点对3种细胞的抑制作用。结果 MTT实验发现总生物碱(LGT总生物碱)及3种倍半萜类生物碱组分(LGT-ZF1、2、3)有细胞增殖抑制活性。其中LGT-ZF1对人CIK细胞、HCT116细胞、HepG2细胞的IC50值分别为(2.08±0.11)μg/mL、(2.36±0.16)μg/mL、(6.05±0.17)μg/mL,抑制效应呈时间剂量依赖性。结论 LGT-ZF1组分具有较好的免疫抑制活性及抗肿瘤作用。 Objective to screen immunosuppressive and antitumor components from Tripterygium wilfordii alkaloids series component,evaluation of toxicity reduction effects of its efficiency. Methods using the MTT method,the total alkaloids of Tripterygium Wilfordii and sesquiterpene alkaloids were 5 components on human CIK cells,HCT116 cells of colorectal adenocarcinoma and hepatocellular carcinoma HepG2 cells,and LGT-ZF1group selected sub inhibitory effects on 3 kinds of cells at different time points. Results the results of MTT test found that total alkaloids( LGT total alkaloids) and 3 kinds of sesquiterpene alkaloids components( LGT-ZF1,2,3) inhibited cell proliferation. The LGT-ZF1 on human CIK cells,HCT116 cells,HepG2 cells and IC50 values were( 2. 08 ± 0. 11) μg / mL,( 2. 36 ± 0. 16) μg / mL,( 6. 05 ± 0. 17) μg / mL,inhibitory effect was in a time and dose dependent. Conclusion LGT-ZF1 component has better immunosuppressive activity and antitumor effect.
