Simultaneous Determination of Protocatechuic Acid, Catechin and Epicatechin in Caulis Spatholobi by Fast Liquid Chromatography

摘要 【目的】建立快速液相色谱法同时测定鸡血藤药材中原儿茶酸、儿茶素、表儿茶素含量。【方法】采用Phenomenex Kinetex XB-C18(4.6 mm×100 mm,2.6μm)色谱柱,以乙腈—磷酸—水为流动相,等度洗脱(体积比为10∶90),流速1.3mL/min,柱温25℃,检测波长260 nm,进样体积2μL。【结果】原儿茶酸、儿茶素、表儿茶素的线性范围分别0.091 6~0.549 6μg(r=0.999 7)、0.772~3.86μg(r=0.999 4)、0.670 5~4.023μg(r=0.999 7);平均加样回收率分别为102.96%(sR=1.62%)、99.44%(sR=2.68%)、97.34%(sR=1.88%)。【结论】该方法简便、快捷,可用于鸡血藤中原儿茶酸、儿茶素、表儿茶素的含量测定,可为鸡血藤的质量控制研究提供参考。 Objective To establish a method for simultaneous determination of protocatechuic acid, catechin and epicatechin in Caulis Spatholobi by fast liquid chromatography. Methods The fast liquid chromatography was performed on Phenomenex Kinetex XB-C18column(4.6 mm×100 mm, 2.6 μm) with acetonitrile-acetic acidwater as mobile phase by isocratic elution(10∶90). The flow rate was 1.3 mL / min, the column temperature was 25℃, the detection wavelength was 260 nm, and the injection volume was 2 μL. Results The linearity of protocatechuic acid, catechin and epicatechin was in the range of 0.091 6~ 0.549 6 μg(r=0.999 7), 0.772 ~ 3.86 μg(r=0.999 4) and 0.670 5 ~ 4.023 μg(r=0.999 7), respectively, and their average recoveries were 102.96%(sR=1.62%), 99.44%(sR=2.68%) and 97.34%(sR=1.88%). Conclusion The established method is simple and fast for the determination of protocatechuic acid, catechin and epicatechin in Caulis Spatholobi, which will supply reference for the quality control of Caulis Spatholobi.
