Effect of up-regulation of NOD2 on biological behavior of human decidual stromal cells during the first trimester of pregnancy

摘要 目的:探讨胞内模式识别受体NOD2表达上调对于人早期妊娠蜕膜基质细胞基本生物学行为的影响。方法:分离培养人早期妊娠蜕膜基质细胞运用CCK8试剂盒、Transwell侵袭小室、流式细胞术等分别检测接受NOD2配体处理后细胞增殖、侵袭和凋亡能力。结果:蜕膜基质细胞接受相应配体刺激后NOD2 mRNA及蛋白表达水平上调,抑制细胞侵袭能力促进细胞凋亡,但是对细胞增殖能力无明显影响。结论:NOD2过度激活损伤蜕膜组织,可能导致不良妊娠结局。 Objective: To explore the effect of NOD2 on biological behavior of human decidual stromal cells during the first trimes- ter of pregnancy. Methods: Human decidual stromal cells during the first trimester of pregnancy were isolated and cultured, then CCK8 kit, Trans - well assay and flow cytometry were used to detect proliferation ability, invasive ability and apoptotic ability of human decidual stromal cells after treated with NOD2 ligand. Results: After stimulated with NOD2 ligand, the expression levels of NOD2 mRNA and protein in human decidual stromal cells were up - regulated, their invasive ability was inhibited and apoptosis was promoted, but it had no significant impact on cell proliferation. Conclusion: Excessive activation of NOD2 can injury decidual tissue and induce adverse pregnancy outcome.
