摘要
目的:通过通顶散各萃取部位对ECV-304的抗过氧化氢损伤作用进行研究,并对其抗过氧化氢损伤活性部位进行化学成分分析。方法:以体外培养的人脐静脉内皮细胞(ECV-304)为研究材料,建立以H2O2诱导内皮细胞的氧化损伤模型,以内皮细胞的存活率为指标,研究通顶散各萃取部位对ECV-304的保护作用;采用高效液相色谱-质谱联用技术对通顶散正丁醇萃取部位化学成分进行分析。结果:通顶散正丁醇萃取部位能够提高ECV-304细胞存活率,具有抵抗H2O2诱导的内皮细胞氧化损伤的作用,经过液质联用分析研究,从通顶散正丁醇萃取部位中共分析推测出20个化学成分。结论:通顶散正丁醇萃取部位为通顶散抗H2O2氧化损伤活性部位。藜芦生物碱、人参皂苷和甘草皂苷是通顶散抗过氧化氢损伤活性的主要物质基础。
Objective: To investigate the protective effect of different solvent fractions of Tongdingsan against damage caused by hydrogen peroxide(H2O2) in human vascular endothelial cells(ECV-304),and analyze fraction of Tongdingsan with the protective activity by HPLC-DAD-ESI-MS / MS. Methods: ECV-304 cells were cultured in vitro,and injured by H2O2. Cell survival was investigated as an indicator to evaluate the protective effect of different solvent fractions of Tongdingsan against damage caused by H2O2. Active Tongdingsan fractions were qualitatively analyzed by HPLC-DAD-MS. Results: n-Butanol fraction of Tongdingsan improved the viability of ECV-304 cells after exposure to H2O2,showing a protective effect on oxidative injury of endothelial cells. Based on determination by UV and mass spectral analysis,a total of 20 compounds were tentatively characterized. Conclusion: The active fraction of Tongdingsan is the n-butanol extract that protects ECV-304 cells against damagecaused by H2O2. Veratrine,ginsenosides and glycyrrhiza uralensis saponins are the main antioxidative substances from Tongdingsan.
