摘要 利用廉价的麸皮作为培养基的碳源,采用单因素试验分析了发酵影响因素摇床转速、发酵温度、接种量、初始pH、发酵时间对Penicillium sp.1407产纤维素酶的影响;利用Box-Behnken试验设计及响应面分析法进行了优化,确定最佳发酵条件为:发酵温度32℃、发酵时间142 h、初始pH 5.2、转速230 r/min,纤维素酶活力为(136±0.74)IU/mL,验证试验结果与模型预测相符。 With the inexpensive wheat straw as a carbon source , the factors affecting cellulase fermentation produced by Penicillium sp. 1407 including shaker's rotate speed ,fermentation temperature, inoculum volume, the initial pH value, fermentation time were analyzed through the monofactorial experiment. Then the Box -Behnken experimental design and response surface methodology (RSM) were taken on regression analysis. The experimental results showed that the optimum fermentation conditions for Penicillium sp. 1407 were fermentation temperature of 32 ℃, fermentation time of 142 h,the initial pH value of 5.2, shaker's rotate speed of 230 r/rain and the cellulase activity reached (136 ±0.74) IU/mL ,which was consistent with the model predicted value.
