摘要 目的探讨1-甲基乙内酰脲对人结肠癌SW480细胞增殖的抑制作用。方法以体外培养的人结肠癌SW480细胞株为实验研究对象。实验分为对照组(等量PBS)和1-甲基乙内酰脲组(M)。MTT比色法检测细胞增殖抑制率,1-甲基乙内酰脲对SW480细胞增殖的抑制作用:根据文献数据和预实验,将1-甲基乙内酰脲组设8个浓度组(M:0.5 mg/L、5 mg/L、10 mg/L、20 mg/L、50 mg/L、100 mg/L、200 mg/L、500 mg/L);1-甲基乙内酰脲八个浓度组分别干预人结肠癌SW480细胞24、48及72 h后,MTT法检测1-甲基乙内酰脲对SW480细胞增殖的抑制作用。结果 MTT法检测结果显示:①1-甲基乙内酰脲在5~500 mg/L浓度范围能抑制SW480细胞增殖,增殖抑制率呈浓度递增趋势。各组间两两比较差异有显著性(P〈0.05)。②1-甲基乙内酰脲作用于结肠癌SW480细胞48 h后,对SW480细胞增殖抑制作用最强。结论 1-甲基乙内酰脲在体外对人结肠癌SW480细胞有抑制增殖的作用。 [ Objectives] To analysis the inhibit proliferation and induced apoptosis effects of 1-Methylhydantoin in human colonic cancer cell line SW480. [ Methods ] Cultivated human colon cancer SW480 cells in vitro as the study abject, MTT assay was used to detect the ahsorbance after treatment with 1-Methylhydantoin at different con- centrations (0, 0.5, 5, 10, 20, 50, 100, 200, 500 rag/L) alone or in combination with 5-fluorouracil at different con- centrations (10, 20, 50, 100 mg/L) on the proliferation of SW480 cells for different time (24 h, 48 h, and 72 h), then the inhibition ratio on SW480 cells was derived. 1-Methylhydantoin and 5-fluorouraci with a special concentration (100 rag/L) respectively effect on SW480 cells ordinary microscope. [Results ] MTF assay results of proliferation in- hibitory rate in experimental groups show that: 1-Methylbydanto could suppress proliferation of SW480 cells within the range of experimental concentrations(0.5, 5, 10, 20, 50, 100, 200, 500 mg/L), and the function intension depend- ed on the density of 1-Methylhydantoin (P 〈0.05). 1-Methylhydantoin exhibited a highest inhibition ratio on colonic cancer cell line SW480 after effecting 48 hour. [ Conclusions ] 1-Methylhydantoin could inhibit proliferation and in- duced apoptosis of colon cancer SW480 cells in vitro, blocked cell cycle in G0/G1 phase.
