基底细胞癌、鳞状细胞癌组织中PinX1、hTERT的表达及意义

摘要 目的分析基底细胞癌、鳞状细胞癌组织中PinX1及hTERT的表达,探讨PinX1、hTERT在基底细胞癌、鳞状细胞癌发生、发展中的作用。方法应用实时定量PCR检测13例基底细胞癌、22例鳞状细胞癌组织中PinX1、hTERT mRNA表达水平,应用免疫组化SP法检测PinX1蛋白和hTERT蛋白在30例基底细胞癌、46例鳞状细胞癌组织中的表达。结果基底细胞癌、鳞状细胞癌组织中PinX1 mRNA表达水平均低于正常组,基底细胞癌、鳞状细胞癌组织中hTERT mRNA表达水平均明显高于正常组,PinX1蛋白在基底细胞癌、鳞状细胞癌组织中的阳性表达率明显低于正常组,hTERT蛋白在基底细胞癌、鳞状细胞癌组织中的阳性表达率明显高于正常组,差异均有统计学意义(P<0.05);基底细胞癌、鳞状细胞癌组织中PinX1蛋白和hTERT蛋白的阳性表达呈负相关(r=-0.460,P<0.05)。结论 PinX1和hTERT参与了基底细胞癌、鳞状细胞癌的发生、发展,二者在基底细胞癌、鳞状细胞癌细胞凋亡方面可能存在拮抗作用。 Objective To observe the expression of PinX1 and hTERT in the basal cell carcinoma (BCC) and squamous cell carcinoma (SCC), and to explore the relationship of PinX1 and hTERT expression with BCC and SCC. Methods Expression of PinX1 and hTERT was detected with real - time quantitative PCR in 13 cases of BCC , 22 cases of SCC. The expression of PIN2/TERF1 interacting telomerase inhibitor 1 ( PinX1 ) and human telomerase reverse transcriptase (hTERT) protein in 30 cases of BCC, 46 cases of SCC were detected with im- munohistochemistry . Results The expression of PinX1 mRNA in tissue of BCC and SCC groups were lower than control group (P 〈 0.05 ) ; the expression of hTERT mRNA in tissue of BCC and SCC groups were significantly higher than control group (P 〈 0.05 ) ; The positive rate of PinX1 in BCC and SCC groups were significantly low- er than that in normal group ( P 〈 0. 05 ) ; The positive rate of hTERT in BCC and SCC group were significantly higher than that in normal group ( P 〈 0.05 ) ; There was a statistically negative correlation between PinX1 and hTERT in BCC and SCC ( r = - 0. 460, P 〈 0.05 ). Conclusion PinX1 and hTERT maybe play an important role in the development of BCC and SCC. The potential mechanisms appear to be that PinX1 and hTERT have antagonistic effects in the process of apoptosis.
