鹅FSHβ基因克隆及序列分析

摘要 促卵泡激素β亚基(FSHβ)基因是动物繁殖性状的一个重要候选基因,对鹅产蛋量具有重要的调节作用。本研究以鹅垂体组织总RNA反转录的cDNA为模板,分别采用RT-PCR和RACE技术扩增鹅FSHβcDNA序列,包含:16 bp 5′UTR,396 bp开放阅读框(ORF)和3′端非翻译区2个不同的剪接体,其大小分别为518 bp和780 bp。经生物信息学软件分析,发现鹅FSHβ基因编码的蛋白为亲水蛋白,且存在信号肽序列切割位点,则推测其为分泌蛋白。本研究首次成功获得了鹅FSHβ基因的cDNA全长序列并进行了分析,可为全面解析鹅FSHβ基因的分子结构及功能提供一定的参考。 Follicle-stimulating hormone beta subunit (FSHfl)gene is an important candidate gene for animal reproductive traits, and also plays a key role in the regulation of egg laying in goose. In this study, FSHfl gene from pituitary gland of goose was cloned by reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA end (RACE) method. The results showed that the cDNA of gFSHβ contained a 16bp 5'UTR,a 396 bp open reading frame and alternative 3'UTRs (518 bp and 780 bp). Bioinformatics analysis suggested that FSHβ protein was hydrophilic and secreted with cleavage sites for signal peptide. The results acquired the gFSHβ cDNA sequence for the first time, which could provide consultation for analysis of the molecular structure and function of FSHfl gene in goose for further study.
