摘要
采用微波提取与分子荧光分析技术,探索正交试验与改进单纯形试验设计相结合优化花生根中白藜芦醇的微波提取工艺.通过L9(34)正交试验考察料液比、微波功率、乙醇溶剂浓度以及微波提取时间等因素对白藜芦醇提取效果的影响,再采用改进单纯形试验进一步优化提取工艺.研究表明:结合正交试验与改进单纯形法提取花生根中白藜芦醇的最佳工艺条件为料液比1∶15(g·mL-1),微波功率中高火,微波时间5 min,乙醇浓度68%.在最佳工艺条件下白藜芦醇的平均提取率为0.151 5%,与单纯采用正交试验最优提取条件相比白藜芦醇的提取率提高了24.30%,有效地实现了花生根中白藜芦醇的最佳提取效果.
By microwave extraction and molecular fluorescence analysis, a new microwave extraction process condition of resveratrol in root of peanut is made through combining with orthogonal test and the improved simplex method. Adopting the method of orthogonal test with L9 (3^4), the influencing factors of extraction conditions for extracting resveratrol in root of peanut were discussed, such as solid-liquid ratio, microwave temperature, concentration of ethanol solvent and microwave time. And then further optimization of extraction process with the improved simplex test. The results show that the optimum technology conditions for extracting resveratrol in root of peanut by microwave extraction method and improved simplex method are as follows: the ratio of material to liquid was 1:15 (g·mL^-l), microwave power of middle high fire, extraction time of 5 rain, volume fraction of ethanol was 68%. With these conditions, the extraction rate of resveratrol from the root of peanut is 0.151 5%. Compared with only use orthogonal experiment, the extraction rate is increased by 24.30%. It can obtain the best extraction conditions of resveratrol from the rood of peanut.
