人体胃癌组织中Smad3基因与Smad7基因的相关性表达及其临床意义 被引量:3

Correlation between Smad3 gene and Smad7 gene in human gastric carcinoma and its clinical significance

摘要 目的:研究胃癌组织中Smad3基因与Smad7基因的表达水平以及二者的相关性,并探索其临床意义。方法:采用免疫组织化学和RT-PCR的方法,检测50例胃癌,癌旁组织以及10例正常胃壁组织中,Smad3与Smad7,蛋白与mRNA的表达。结果:胃癌组织中Smad3蛋白表达阳性率明显低于癌旁组织(50.0%vs.92.0%,P=0.000),2组相比有统计学差异。并与肿瘤的分期(P=0.004)及肿瘤组织的分化(P=0.030)密切相关。而与年龄(χ2=0.439,P=0.508)、性别(χ2=0.117,P=0.733)均无关,胃癌组织中Smad7蛋白的表达阳性率高于癌旁组织(48.0%vs.4.0%,P=0.000),正常组织最低。且与肿瘤的分期(P=0.009),组织的分化程度(P=0.001)密切相关。在胃癌组织中发现Smad3蛋白与Smad7蛋白呈负相关性(r=-0.539,P=0.000)。Smad3 mRNA在癌组织中的表达低于癌旁组织(Z=7.84,P=0.000),而Smad7 mRNA在癌组织中的表达高于癌旁组织(Z=7.83,P=0.000)。结论:Smad3基因在癌组织中低表达,而Smad7基因在癌组织中则高表达,并且二者与胃癌的分期,组织分化程度有关,提示二者在胃癌的发生、发展中有一定的作用,可能和胃癌的分化,分期有关。 Objective:To explore the expression of Smad3 gene and smad7 gene in human gastric carcinoma,their correlation and clinical signifieances. Methods:Totally 50 cases of gastric carcinoma with adjacent tissues and 10 cases of normal gastric tissues were collected in this study. Smad3 and Smad7 protein and mRNA expression in these samples was detected by immunohistochemistry and RT-PCR. Results:Smad3 protein expressions in gastric carcinoma tissue was significantly lower than that in the adjacent carcinoma tissue(50.0% vs. 92.0% ,P=0.000). The expression was closely related with tumor stage(P=0.004) and tumor differentiation stage (P=0.030). The expression was not related with patients' age (c=0.439,P=0.508)or gender (X2=0.117, P=0.733). Positive rate of Smad7 protein expression in gastric carcinoma tissue was higher than that in adjacent carcinoma tissue(48.0% vs. 4.0%,P=0.000) with that in normal tissue being the lowest. The positive rate was closely related with tumor stage(P=0.009) and degree of differentiation (P=-0.001). Smad3 protein and Smad7 protein was found negative correlated in gastric carcinoma(r=-0.539,P=0.000). Smad3 mRNA expression in carcinoma tissue was lower than that in adjacent carcinoma tissue (Z=7.84, P=0.000), but Smad7 mRNA expression in carcinoma tissue were higher than those in adjacent carcinoma tissue(Z=7.83 ,P=0.000). Conclusions:Smad3 gene expression in carcinoma tissue is decreased while Smad7 gene expression in cancer tissue is increased. Smad3 gene expression and Smad7 gene expression are related with stage of gastric carcinoma and degree of differentiation. Correlated expression of these two genes plays certain role in the occurrence and development of gastric carcinoma.
