胃癌组织EZH2与凋亡相关蛋白Caspase-3、Bcl-2、Bax表达的关系及意义 被引量:7

摘要 目的探讨胃癌组织中果蝇zeste基因增强子同源物2(EZH2)、Caspase-3、Bcl-2、Bax的表达关系及其临床意义。方法收集62例胃癌手术患者的肿瘤及52例癌旁正常胃黏膜组织石蜡标本进行免疫组织化学(SP)染色,同时检测凋亡相关蛋白Caspase-3、Bcl-2、Bax蛋白的表达,相关分析探讨EZH2与这3种蛋白质之间的关系及意义。结果胃癌组织中EZH2、Bcl-2表达与癌旁正常组织比较明显增强(P均<0.05),Caspase-3、Bax蛋白在胃癌组织中表达较癌旁组织明显降低(P均<0.05);EZH2蛋白表达随肿瘤分化程度降低而升高、随分期的升高而升高;相关分析显示,EZH2与Caspase-3、Bcl-2与Bax表达存在负相关关系(P均<0.05);其余蛋白之间表达无明显相关性(P>0.05)。结论 EZH2蛋白在胃癌组织中表达增强,且与肿瘤分化程度及分期有关;EZH2蛋白参与胃癌的发病机制在于其可能通过抑制Caspase-3表达而增强胃癌细胞抗凋亡能力。 Objective It is to investigate the relationship and clinical significance between the expression of EZH2, Caspase -3,Bcl-2,Bax in gastric cancer. Methods The expressions of EZH2,Caspase-3,Bcl-2 and Bax were tested by immunohistochemically in 62 tumor tissue specimens of gastric cancer and 52 cases of tumor-adjacent tissues. The relationship and significance of these proteins were analyzed. Results Positive rates of EZH2,Bcl -2 in tumor tissues were higher than those in tumor-adjacent tissues,and expressions of Caspase-3,Bax were lower in tumor tissues than in tumor-adjacent tissues ( all P〈0. 05). The expression of EZH2 was positively related to degree of differentiation and TNM stages(all P〈0. 05). The expression of EZH2 was negatively correlated to Caspase-3,Bcl-2 and Bax(both P〈0. 05). Conclusion EZH2 is over ex-pressed in gastric cancer,and expression of EZH2 is related to differentiation and TNM stages. EZH2 may promote ability of apoptosis resistance of tumor cells by regulating expressions of Caspase-3.
