红法夫酵母原生质体制备条件优化及紫外诱变高产虾青素菌株筛选 被引量:2

Optimization of preparation conditions of Phaffia rhodozyma protoplasts and screening of astaxanthin-yielding strains by ultraviolet mutagenesis

摘要 采用单因素试验和正交试验对红法夫酵母原生质体制备条件进行优化,并用紫外诱变的方法筛选虾青素高产菌株。结果表明:菌龄18h、蜗牛酶浓度1%、30℃处理2h,在以蔗糖为渗透压稳定剂的情况下较有利于原生质体的形成与再生;在70%~85%的致死率条件下筛选到1株高产诱变菌株,虾青素产量较出发菌株提高85.6%。 The preparative conditions of Phaffia rhodozyma protoplasts were optimized by single factor test and orthogonal test and the strains having a high astaxanthin yield were selected by ultraviolet mutagenesis. The results showed that the favourable conditions for the protoplasts to form and regenerate were the cell age of 18 h,helicase concentration of 1% ,treatment at 30 ℃ for 2 h,and osmotic pressure stabilizer of 0.5 mol/L sucrose;A mutant strain whose astaxanthin yield was 85.6% higher than its initial strain was selected under 70% - 85% of fatality rate.
