魔芋内生拮抗菌的定殖研究 被引量:2

摘要 采用抗魔芋(Amorphophallus rivieri Durieu)软腐病的内生芽孢杆菌BS-8发酵液灌根魔芋植株,定期测定该内生拮抗菌在魔芋叶片中的定殖情况。根据该内生菌16S rRNA特异性保守区域设计引物,定期4次采样,提取魔芋叶片DNA,通过SYBR Green I实时荧光定量PCR法检测该内生拮抗菌在魔芋叶片中的定殖情况。结果表明,经内生拮抗菌灌根培养的魔芋叶片中芽孢杆菌数量均明显大于对照,该内生芽孢杆菌能大量定殖在魔芋叶片中。 The colonization condition of an antagonistic and endophytic bacterium named BS-8 resistant to konjac(Amorphophallus rivieri Durieu) soft rot was determined through irrigating the fermentation broth of BS-8 to the root of konjac plants. With the specificity of conserved domain of BS-8 16S rRNA,several primers were designed to amplify the fragment of the target gene. The leaves of konjac plants were sampled four times regularly. With the DNA extracted from the samples, the colonization condition of BS-8 in konjac leaves was tested by SYBR Green I real-time fluorescence quantitative PCR. The results showed that the quantity of Bacillus bacteria in the leaves irrigated with fermentation broth was significantly higher than the contrast. The conclusion above suggests that BS-8 can largely colonize in konjac leaves.
